芹菜素对急性大鼠脑缺血再灌注后脑组织NCAM表达的影响

【摘要】:目的:探讨NCAM在大鼠脑缺血再灌注后不同时间的表达变化及芹菜素对其表达的影响。方法:采用改良线栓法建立大鼠脑缺血再灌注后不同时间损伤模型。动物随机分为假手术组(S)、模型组(M)、芹菜素组(A)和地塞米松组(D)。各手术组按再灌注时间不同又分为再灌注6 h组、24 h组、72 h组、7 d组,各组动物麻醉清醒后均给予神经行为学评分,每组1只大鼠TTC染色观察脑梗死灶、6只免疫组化法测定NCAM的表达。结果在A72h组神经行为学评分低于M72h(P0.05);脑组织切片TTC染色显示各手术组均出现白色梗死灶;正常脑组织NCAM少量表达,缺血再灌注后表达增加,于72 h达高峰,A组能显著提高NCAM的表达,各时间点A和M组比较差异显著(P0.05)。结论:芹菜素可增加脑缺血再灌注后NCAM的表达,提示芹菜素可能通过NCMA达到促进脑组织损伤后神经细胞修复和再生的作用。
[Abstract]:Aim: to investigate the changes of NCAM expression at different time after cerebral ischemia reperfusion in rats and the effect of apigenin on it. Methods: the rat model of cerebral ischemia reperfusion injury at different time was established by modified thread embolization. Animals were randomly divided into sham operation group (S), model group (M), apigenin group (A) and dexamethasone group (D). Each operation group was divided into 6 h group, 24 h group, 72 h group and 7 day group according to the time of reperfusion. The rats in each group were given neurobehavioral score after anaesthesia. The cerebral infarction was observed by TTC staining in each group. The expression of NCAM was detected by immunohistochemical method in 6 mice. Results the neurobehavioral score in A72 h group was lower than that in M72 h group (P0.05), and the white infarct foci appeared in all the operation groups by TTC staining. The expression of NCAM in normal brain tissue increased after ischemia reperfusion and reached the peak at 72 hours. Group A could significantly increase the expression of NCAM, and the difference was significant between group A and group M at each time point (P0.05). Conclusion: apigenin can increase the expression of NCAM after cerebral ischemia-reperfusion, suggesting that apigenin can promote the repair and regeneration of nerve cells after brain injury through NCMA.
