七氟烷麻醉大鼠血中游离六氟异丙醇浓度测定

【摘要】:目的:测定成年雄性SD大鼠经不同浓度七氟烷麻醉不同时间后血中游离六氟异丙醇(HFIP)的浓度,并分析游离HFIP含量与七氟烷麻醉浓度、麻醉时间之间的关系。方法:取健康成年雄性SD大鼠100只,随机分为四组,每组25只,分别接受0.5MAC、1MAC、1.5MAC、2MAC浓度的七氟烷麻醉。七氟烷麻醉气体由麻醉机上的挥发罐输向大鼠专用麻醉装置。麻醉机输出七氟烷的实际浓度经气相色谱仪进行校准。各组大鼠分别麻醉0.5h、1h、2h、4h、6h后取其动脉血7毫升,用二次平衡法测定血中游离六氟异丙醇浓度。求出每组数据平均值,分别绘制“吸入七氟烷浓度-血中游离六氟异丙醇浓度”关系曲线及“麻醉时间-血中游离六氟异丙醇浓度”关系曲线,分析HFIP含量与七氟烷浓度、麻醉时间之间的关系。结果:各组SD大鼠经七氟烷麻醉后血中均能检测出游离HFIP。组内比较:各组大鼠血中游离HFIP浓度随时间变化趋势基本一致,即0.5h至4h间HFIP浓度随麻醉时间延长而逐渐增加,HFIP峰值出现在接受七氟烷麻醉后第四小时,在第4至第6小时间呈下降趋势;组间比较:0.5MAC组各时点游离HFIP浓度为四组中最低者;而1.0MAC和1.5MAC组各时点游离HFIP浓度均较高,且都明显高于2.0MAC组,但1.5MAC组血中游离HFIP随麻醉时间变化趋势最为平稳。结论:血中游离HFIP浓度随七氟烷麻醉时间的延长呈现先增高而后降低的趋势;中等浓度七氟烷麻醉组血中游离HFIP浓度水平高于低浓度麻醉组,但继续增大七氟烷麻醉浓度(2.0MAC),血中所产生的游离HFIP浓度反而降低。
[Abstract]:Aim: to determine the concentration of free hexafluoropropanol (HFIP) in blood of adult male SD rats after sevoflurane anesthesia with different concentrations of sevoflurane, and to analyze the relationship between free HFIP content and sevoflurane anesthetic concentration and anesthetic time. Methods: 100 healthy adult male SD rats were randomly divided into four groups (25 rats in each group). Sevoflurane anesthetic gas is transported from volatile tank on anaesthesia machine to special anesthetic device in rats. The actual concentration of sevoflurane from anaesthesia machine was calibrated by gas chromatograph. Rats in each group were anesthetized for 0. 5 h, 1 h, 2 h and 4 h for 6 h. After 6 hours, 7 ml of arterial blood was taken, and the concentration of free hexafluoropropanol in blood was determined by two times equilibrium method. The average values of each group were calculated, and the curves of "inhaled sevoflurane concentration-blood free hexafluoropropanol concentration" and "anesthetic time-blood free hexafluoropropanol concentration" were drawn, respectively. The HFIP content and sevoflurane concentration were analyzed. The relationship between anesthetic time. Results: free HFIP. could be detected in the blood of SD rats after sevoflurane anesthesia. Comparison within the group: the concentration of free HFIP in blood of rats in each group showed the same trend with time, that is, the concentration of HFIP increased gradually with the prolongation of anesthesia time between 0.5 h and 4 h, and the peak value of HFIP appeared in the fourth hour after sevoflurane anesthesia. From the 4th to the 6th, there was a downward trend. Comparison between groups: free HFIP concentration in 0.5MAC group was the lowest in the four groups at each time point. The concentration of free HFIP in 1.0MAC and 1.5MAC groups was higher than that in 2.0MAC group at each time point, but the trend of free HFIP in 1.5MAC group was the most stable with anesthesia time. Conclusion: with the prolongation of sevoflurane anesthesia time, the concentration of free HFIP in blood increased first and then decreased. The level of free HFIP in the middle concentration sevoflurane anesthesia group was higher than that in the low concentration anesthesia group, but it continued to increase the sevoflurane anesthetic concentration (2.0MAC), but the free HFIP concentration in the blood decreased.
