寒冷刺激对大鼠胃粘膜的影响及良附丸的防治作用研究

【摘要】:目的:观察内、外寒冷刺激对大鼠胃粘膜形态及相关功能的影响。通过对实验大鼠施加寒冷刺激因素,观察大鼠胃粘膜局部病理改变及检测血清和胃粘膜的胃动素、胃泌素、前列腺素及内皮素的变化,探讨内外寒冷刺激对大鼠胃粘膜功能状态的影响,及良附丸的防治作用。 材料与方法: 1分组造模及用药干预:大鼠适应性喂养3天,按体重随机分为5组。正常对照组:用生理盐水灌胃2mL/l00g,日2次;冰水浸泡组:用0℃冰水浸泡大鼠30min,水深为2cm,日1次;生理盐水灌胃2mL/l00g,日1次;冰水灌胃组:用0℃冰水灌胃2mL/100g,日2次;复合刺激组:用0℃冰水浸泡大鼠30min,水深为2cm,用0℃冰水2mL/100g灌胃,日1次;中药防治组:用0℃冰水浸泡大鼠30min,,水深为2cm,用0℃冰水2mL/100g灌胃,日1次,同时给予良附丸溶液灌胃,日1次,灌胃时间间隔4小时以上。实验周期共7天。 2大鼠一般状态的观察:实验进行过程中,对大鼠饮食,二便,精神状态,毛色,体重情况及死亡情况进行观察和记录。 3取材及实验指标检测:实验结束后,禁食不禁水12小时,麻醉后,剖开大鼠腹腔,腹主动脉采血4mL,检测血清胃动素、胃泌素、前列腺素,内皮素;沿大弯侧剪开鼠胃,生理盐水冲洗,取胃体、胃窦组织检测胃粘膜病理,及胃粘膜胃动素、胃泌素、前列腺素,内皮素含量。 结果: 1施加内、外寒冷刺激可损伤大鼠胃粘膜形态。 2大鼠胃组织和血清中的胃动素、胃泌素、6-keto-PGF2α水平降低,内皮素水平升高。 3良附丸可改善大鼠受损的胃粘膜。 4良附丸对模型大鼠血清及胃粘膜中的胃动素、胃泌素、6-keto-PGF2α水平,内皮素的水平有一定调整作用。 结论: 1寒性刺激可以使实验大鼠的胃粘膜造成病理性损伤。 2寒冷刺激可以使大鼠胃粘膜和血清的胃动素、胃泌素、6-keto-PGF2α水平降低;可以使内皮素水平升高。 3冰水浸泡、冰水灌胃及复合因素对胃粘膜损伤的程度是不同的,复合因素损伤作用较重,冰水灌胃次之,冰水浸泡较轻。 4良附丸对寒冷刺激所致胃粘膜的损伤有一定防治作用。
