白金养胃颗粒的药效学研究

【摘要】:目的:1.白金养胃颗粒急性毒性实验2、模拟大鼠幽门结扎型胃溃疡模型,观察白金养胃颗粒是否具有抗胃溃疡作用,并研究白金养胃颗粒抗胃溃疡作用机理:采用热板法、小鼠耳肿胀法进行小鼠镇痛和抗炎实验,观察白金养胃颗粒是否具有镇痛、抗炎作用。方法:1.采用幽门结扎法模拟大鼠胃溃疡模型,分组及给药:将60只大鼠按体重随机分为空白组、模型组、阳性对照药组(三九胃泰颗粒,3.6g· kg-1)和白金养胃颗粒高、中、低剂量组(2.20,1.10,0.55g· kg-1),每组10只。每天给药1次,连续给药5天。2.模型制备:采用大鼠幽门结扎胃溃疡实验法[4]。首先各组鼠需禁食18h,但不禁水,把鼠放到空笼子中,以控制大鼠饿极吃食自己的粪便、垫料,18h后用乙醚麻醉,麻醉后使把大鼠固定在鼠板上,用碘伏或酒精消毒老鼠皮肤,沿腹中线(自剑突下缘)切开老鼠腹壁(注意切开大鼠皮肤时要先轻后重),切口约为2.5cm,用止血钳撑开腹膜,用手指轻轻推左侧肋缘,向上使胃暴露于视线中,找出胃和十二指肠交界处,在胃和十二指肠连接处穿线(线需事先放在酒精里消毒),切记勿伤周围大血管及毛细血管,然后将幽门结扎,结扎后缝合切口,缝合过程中用阿司匹林和酒精消毒,纱布包扎,之后将各组动物均经直肠给药1次,放回鼠笼中,禁食不禁水。15 h后乙醚麻醉大鼠,并于颈总动脉取血[52]。沿切口打开腹腔,沿胃大弯剪开胃部,摘取全胃放到盛有酒精的烧杯中,观察并测量胃溃疡情况,测量胃溃疡点,胃黏膜的损伤长度,将每只大鼠所有的损伤长度加和作为该鼠胃溃疡指数,并给每个粘膜损伤的程度评分,计算每只鼠所有损伤评分的加和,用"分"表示胃溃疡指数[]。观察白金养胃颗粒对减少大鼠溃疡作用、抑制溃疡指数的作用;并监测各组中大鼠的胃液量、胃液酸度,血清中的胃动素、胃泌素含量,以及前列腺素E2(PGE2)、白介素-1(IL-1)、白介素-7(IL-7)的水平,表皮生长因子(EGF)、一氧化氮(NO)的含量。并检测胃组织中生长抑素(SS)水平、丙二醛(MDA)、H+-K+-ATP酶、超氧化物歧化酶(SOD)含量的变化程度。小鼠镇痛实验中(选用热板法),实验前于热板仪上测定痛阂值(第1次添后足的时间作为指标),重复测取2次正常痛阈值作为给药前痛阈值[52]。选择水域的水温在(55℃ ± 0.5℃)时的痛阈值在5~30s内的雌性小鼠50只,分组给药1 h后开始测小鼠痛阈值,按标号分组的小鼠计好痛阈值,之后4h每隔1h测各小鼠的痛阈值。记录好小鼠给药前后痛阈值的大小,保存好完整原始实验数据。采用小鼠耳肿胀法进行小鼠抗炎实验,60只小鼠,雌雄参半,按体重随机分,每组10只小鼠,一共分为5组,分成阳性对照药,也就是三九胃泰组(5.20 g · kg-1),空白组、白金养胃颗粒高、中、低剂量组(3.00,1.5,0.75 g · kg-1),每天给药1次,给药1天。分组给药后于末次给药1 h后,在每只小鼠右耳廓(耳朵两面)涂抹0.02mL二甲苯,以致炎症发生。并在4h后处死小鼠(采用脱颈法),并沿耳廓线剪下两耳,用8mm的打孔器分别在同一部位打下两耳片,并用分析天平称其重量,计算每只小鼠右耳片的质量减去左耳片的质量,就是肿胀程度,并计算抑制率[54]。结论:药效学研究显示,在大鼠幽门结扎型胃溃疡实验中,与模型组比较,白金养胃颗粒三个剂量组均能够显著减小大鼠胃溃疡面积和大鼠胃溃疡指数,可明显减少大鼠胃液量,降低胃酸浓度(p0.01),并且增加血清中PGE2水平,胃泌素、胃动素的的含量(p0.05)。在小鼠镇痛、抗炎实验中,白金养胃颗粒高、中剂量组均能够降低二甲苯所致的小鼠耳肿胀程度(p0.05),白金养胃颗粒各给药组均可不同程度提高小鼠痛阈值(p0.01,p0.05)。白金养胃颗粒可以治疗幽门结扎型胃溃疡疾病,并且白金养胃颗粒还具有镇痛与抗炎作用。
