酸脂清对尿酸钠晶体所致痛风性关节炎模型大鼠软骨破坏的影响

【摘要】:目的:通过研究酸脂清不同提取物对痛风性关节炎模型大鼠血清IL-1β、TNF-α、IL-10、IL-1Ra浓度、关节肿胀和关节滑膜病理变化的影响,,评价酸脂清对痛风性关节炎的抗炎和关节保护作用。 方法:将80只实验用Wistar大鼠(200±20g,雄性),随机分成空白组、模型组、水提酸脂清组、醇提酸脂清组和秋水仙碱组5组,每组16只,空白组和模型组给予生理盐水灌胃,水提酸脂清组、醇提酸脂清组和秋水仙碱组分别给予酸脂清水提物、醇提物和秋水仙碱片混悬液灌胃,第7天灌胃结束后0.5h,测量大鼠右踝关节直径,并选定踝关节外侧后方以尿酸钠晶体混悬液局部注射造成痛风性关节炎模型。造模后9h、12h在各组中随机抽取8只,测量造模踝关节直径;麻醉后下腔静脉取血,Elisa法检测IL-1β、TNF-α、IL-10、IL-1Ra浓度;将造模踝关节取下,做病理切片观察滑膜炎症变化。 结果:①两酸脂清水提取物组较模型组肿胀程度明显降低,有显著性意义(P<0.05);②三个药物组与模型组比较,IL-1β、TNF-α(软骨破坏因子)浓度明显降低、IL-10、IL-1Ra(保护因子)浓度明显升高(P<0.05);③三个药物组关节滑膜炎症与模型组关节滑膜炎症比较,三个药物组滑膜炎症明显轻于模型组;④酸脂清水提组可明显升高IL-10浓度,与其他各组比较有显著性差异(P<0.05)。 结论:酸脂清通过减轻痛风性关节炎模型大鼠关节炎症,降低关节软骨破坏因子IL-1β和TNF-α浓度,升高软骨保护因子IL-10和IL-1Ra的浓度,达到治疗痛风性关节炎,减少关节损伤的目的。其中酸脂清水提物对IL-10的升高效果较明显,故酸脂清水提物效果较好。
[Abstract]:Aim: to study the effects of different extracts of acid Zhiqing on serum IL-1 尾, TNF- 伪, IL-10,IL-1Ra concentration, joint swelling and pathological changes of synovium in rats with gouty arthritis, and to evaluate the anti-inflammatory and joint protective effects of acid Zhiqing on gouty arthritis. Methods: 80 Wistar rats (200g 卤20g, male) were randomly divided into three groups: blank group, model group, water extract group, alcohol extract group and colchicine group, with 16 rats in each group. The blank group and model group were given saline intragastrically, water extract group, alcohol extract group and colchicine group were given acid lipid extract, alcohol extract and colchicine tablet suspension respectively. 0.5 h after the end of gastric administration on the 7th day, the rats in the blank group and the model group were given saline, water extract, alcohol extract and colchicine tablet suspension, respectively, and the rats in the blank group and the model group were given saline, water extract, alcohol extract and colchicine tablet suspension respectively. The diameter of right ankle joint was measured and the model of gouty arthritis was established by local injection of sodium urate crystal suspension. At 9 h and 12 h after modeling, 8 rats were randomly selected from each group to measure the diameter of malleolus joint, the blood was taken from inferior vena cava after anesthesia, the concentrations of IL-1 尾, TNF- 伪 and IL-10,IL-1Ra were detected by Elisa method, and the malleolus joint was removed and pathological section was made to observe the changes of synovitis. Results: (1) the swelling degree of the extract group was significantly lower than that of the model group (P < 0.05), and the concentration of IL-1 尾, TNF- 伪 (cartilage destruction factor) and IL-10,IL-1Ra (protective factor) in the three drug groups were significantly lower than those in the model group (P < 0.05). 3Compared with the model group, the synovitis in the three drug groups was significantly lighter than that in the model group, and the concentration of IL-10 in the acid extract group was significantly higher than that in the other groups (P < 0.05). Conclusion: Zaozhiqing can treat gouty arthritis and reduce joint injury by reducing arthritis, decreasing the concentration of IL-1 尾 and TNF- 伪, and increasing the concentration of cartilage protective factor IL-10 and IL-1Ra. The effect of acid-fat clear water extract on the increase of IL-10 was obvious, so the effect of acid-lipid clean water extract was better.
