刺五加皂苷对急性脊髓损伤后脊髓内BDNF和NGF表达的影响

【摘要】:目的研究刺五加皂苷对脊髓损伤后组织内脑源性神经营养因子(BDNF)和脊髓内神经生长因子(NGF)两种蛋白表达的影响,探讨刺五加皂苷对损伤脊髓的保护作用。方法将45只雄性大鼠随机分为3组:假手术组、模型组和刺五加皂苷干预组,每组15只。将实验动物用10%水合氯醛麻醉后,用改良Allen’s法进行急性脊髓损伤模型造模。造模后,假手术组予正常饲养,模型组予连续7d生理盐水腹腔注射,刺五加皂苷干预组进行连续7d100mg/kg刺五加皂苷持续腹腔注射。ELISA法检测血清中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,通过病理切片分析大鼠脊髓组织病理变化,蛋白质印迹法检测大鼠脊髓内BDNF和NGF表达。结果与假手术组比较,模型组和刺五加皂苷干预组脊髓有明显损伤,且大鼠血清中MDA含量增加(P0.05),SOD活性降低(P0.05),大鼠脊髓组织中的BDNF和NGF蛋白表达量上升(P0.05)。与模型组相比,刺五加皂苷组大鼠脊髓损伤明显改善,血清中MDA含量降低(P0.05),SOD活性升高(P0.05),大鼠脊髓组织中的BDNF和NGF蛋白表达量上升(P0.05)。结论刺五加皂苷促进脊髓组织中神经元细胞内BDNF和NGF表达,利于损伤神经元的修复,对大鼠急性损伤的脊髓具有保护作用。
[Abstract]:Objective to study the effect of Acanthopanax senticosus saponins on the expression of brain-derived neurotrophic factor (BDNF) and spinal cord nerve growth factor (NGF) after spinal cord injury, and to explore the protective effect of Acanthopanax senticosus saponins on spinal cord injury. Methods 45 male rats were randomly divided into three groups: sham operation group, model group and Acanthopanax senticosus saponins intervention group, with 15 rats in each group. The experimental animals were anesthetized with 10% chloral hydrate and the model of acute spinal cord injury was established by modified Allen' s method. After the model was established, the model group was fed with normal saline for 7 days, and the intervention group was treated with continuous intraabdominal injection of Acanthopanax senticosus saponins. The content of malondialdehyde (MDA) and the activity of SOD (SOD) in serum were detected by Elisa, the pathological changes of spinal cord tissue were analyzed by pathological section, and the expression of BDNF and NGF in spinal cord of rats was detected by Western imprinting. Results compared with the pseudo-operation group, the spinal cord injury was obvious in the model group and Acanthopanax senticosus saponins intervention group, and the content of MDA in serum was increased (P 0.05), and the expression of BDNF and NGF protein in spinal cord tissue was increased (P 0.05). Compared with the model group, the spinal cord injury of rats in Acanthopanax senticosus saponins group was significantly improved, the content of MDA in serum was decreased (P 0.05), and the expression of BDNF and NGF protein in spinal cord tissue was increased (P 0.05). Conclusion Acanthopanax senticosus saponins promote the expression of BDNF and NGF in spinal cord tissue, which is beneficial to the repair of injured neurons and has protective effect on acute spinal cord injury in rats.
