TGF-β介导盆腔淋巴管放射性纤维化作用机制的研究
[Abstract]:Objective: to investigate the mechanism of radiation fibrosis of pelvic lymphoid vessels mediated by transforming growth factor-尾 (TGF- 尾). Methods: twenty female New Zealand white rabbits were randomly divided into blank control group (Sham group), 4 weeks after irradiation group, 12 weeks after irradiation and 16 weeks after irradiation. Except Sham group, all the rabbits in the other groups were treated with single radiation and high dose irradiation. At the 4th week, 12th week and 16th weekend, the rabbits in each group were anesthetized with chloraldehyde hydrate. The pelvic lymphatic vessels were dissected and the pathological changes of pelvic lymphoid vessels were observed under staining light microscope. The expression of TGF- 尾, hypoxia inducible factor-1 伪 (HIF-1 伪) and vascular endothelial growth factor (VEGF) were detected by Western imprinting. Results: in Sham group, the cells in pelvic lymphoid tissue of rabbits were neatly arranged and no inflammatory cells infiltrated, a small amount of inflammatory cell infiltration and edema were observed at 4 weeks after irradiation, and red blood cells and macrophages were aggregated at 12 weeks after irradiation, a large number of inflammatory cells infiltrated and edema was significant. At 16 weeks after irradiation, the wall thickened, inflammatory cells infiltrated obviously, collagen proliferation was obvious. Azan staining Sham group showed no fibrosis expression in pelvic lymphoid tissue, blue fibrosis expression was rare in pelvic lymphoid tissue of rabbits 4 weeks after irradiation, and a large number of fibrosis expression was found in rectal tissue 12 weeks and 16 weeks after irradiation. The expression of TGF- 尾 protein in pelvic lymphoid tissue increased with the prolongation of irradiation time, and the expression of HIF-1 伪 protein in nucleus and VEGF protein in cytoplasm also increased with the prolongation of irradiation time. Conclusion: the increase of TGF- 尾 protein expression by irradiation may be related to HIF-1 伪 and VEGF protein. TGF- 尾 may regulate pelvic lymphatic radiation fibrosis by affecting HIF-1 伪.
