丙泊酚对小鼠小脑浦肯野细胞活动及感觉信息传递的影响

丙泊酚对小脑皮层浦肯野细胞自发性活动的影响[目的]丙泊酚,2,6-双(1-甲基乙基)苯酚乳剂,是一种快速静脉镇静催眠药,广泛用于麻醉的诱导与维持,也用于特护病房(ICU)患者的镇静。丙泊酚对整个中枢神经系统均有抑制作用,其最常见副作用是剂量依存地抑制心肺功能。已证实丙泊酚具有活化γ氨基丁酸(GABA)A受体、抑制N-甲基-D-天门冬氨酸(NMDA)受体,并通过慢速钙离子通道调节钙内流。在离体条件下,丙泊酚可通过活化GABAA和甘氨酸受体对脊髓、海马和下丘脑神经元的活动产生抑制作用,然而,丙泊酚对活体动物小脑皮层神经元活动的影响尚不清楚。因此,我们应用细胞贴附式记录和药理学手段研究丙泊酚对乌拉坦麻醉小鼠浦肯野细胞自发性活动的影响,旨在阐明静脉麻醉药丙泊酚对小脑皮层浦肯野细胞自发性单纯性放电和复杂性放电的影响机制。[方法]成年ICR小鼠(6-8周龄)通过腹腔注射乌拉坦麻醉(1.3g/kg体重)后,行气管插管以保证呼吸道畅通,然后将动物固定在自制的脑立体定位仪上。在小脑部位制作一个灌流槽后,在小脑CrusⅡ区相对应处行开颅手术,钻一个直径约为1-1.5mm小孔,暴露记录部位的小脑表面,轻轻去...
