丙泊酚对小鼠小脑皮层分子层和颗粒细胞层感觉信息传递的影响

  [目的]丙泊酚是一种中枢神经系统抑制剂,在体内条件下剂量依赖性的抑制大脑新皮层的信息处理。然而,丙泊酚对活体动物小脑皮质分子层感觉信息处理的影响还不清楚。因此,我们在乌拉坦麻醉麻醉下,应用电生理学和药理学方法研究丙泊酚对感觉刺激诱发小鼠小脑分子层场电位反应的影响。[方法]6-8周龄的成年ICR小鼠通过腹腔注射体重为1.3g/kg乌拉坦麻醉后,进行气管插管以避免呼吸道堵塞,然后将小鼠固定在定制的立体定位装置上。在小脑相应部位作一个防水外流灌流槽后,在小脑Crus Ⅱ区的相对应部位行开颅手术,钻一个直径为1-1.5mm的小孔,暴露记录部位的小脑表面,并用蠕动泵在暴露脑表面持续灌流含氧的人工脑脊液(artificial cerebrospinal fluid, ACSF)o使用Axopatch-200B膜片钳放大器(Molecular Devices, Foster City, CA)记录感觉刺激诱发小脑皮层分子层场电位,并通过D/A转换器1440和Clampex 10.3软件获取感觉刺激诱发场电位活动的数据。记录电极内填充的是人工脑脊液,阻抗在3-5 MΩ之间。丙泊酚和gabazine购置...
