右美托咪定通过p38通路改善发育期大鼠七氟醚麻醉后认知功能障碍

目的:探讨右美托咪定通过p38通路改善发育期大鼠七氟醚麻醉后认知功能障碍。方法:将90只发育期大鼠根据随机数字法分为对照组（C组）、七氟醚组（S组）、七氟醚+右美托咪定组（S+D组）,每组30只。缺口末端标记法（TUNEL）染色测定大鼠海马CA1区凋亡细胞数,免疫荧光染色测定大鼠海马CA1区Caspase-3、B淋巴细胞瘤-2基因（Bcl-2）及相关的蛋白（Bax）表达,免疫组化染色测定大鼠海马CA1区p38丝裂原活化蛋白激酶（p38MAPK）表达,采用Western blot测定海马CA1区p38MAPK、磷酸化p38MAPK（p-p38MAPK）蛋白水平,Morris水迷宫实验测定认知功能。结果:与C组比较,S组和S+D组大鼠海马CA1区凋亡细胞数升高（P<0.05）,海马Caspase-3、Bax阳性细胞数和荧光强度升高（P<0.05）,Bcl-2阳性细胞数和荧光强度降低（P<0.05）,海马CA1区p38MAPK免疫组化IHS评分升高（P<0.05）,海马CA1区p-p38MAPK蛋白水平升高（P<0.05）,逃避潜伏期延长（P<0.05）,...
