【摘要】:目的:探讨MDR1、SLCO1B3基因多态性对肾移植术后他克莫司疗效的影响。 方法:选择中南大学湘雅三医院亲属肾移植术后3个月至6个月稳定期患者,男女不限,年龄18岁-65岁。术后采用他克莫司+霉酚酸酯+皮质类固醇激素三联免疫抑制治疗,EMIT方法检测他克莫司的全血浓度后计算出浓度/剂量(C/D)比值。采用PCR-RFLP对MDR1基因343CT、SLCO1B3基因699GA进行基因分型。比较MDR1基因3435CT、LCO1B3基因699GA不同基因型对他克莫司浓度/剂量(C/D)比值的影响。 结果:MDR1基因3435CT位点不同基因型术后他克莫司C/D比值差异无统计学意义(P0.05)、SLCO1B3基因699GA位点不同基因型术后他克莫司C/D比值差异有统计学意义(P0.05)。 结论:SLCO1B3基因699GA不同基因型对术后他克莫司C/D比值有影响,突变型纯合子AA术后他克莫司C/D比值较高。
[Abstract]:Objective: to investigate the effect of MDR1 / SLCO1B3 gene polymorphism on the efficacy of tacrolimus after renal transplantation. Methods: male and female patients with stable renal transplantation from 3 months to 6 months after renal transplantation in Xiangya third Hospital of Central South University were selected. After operation, the whole blood concentration of tacrolimus was measured by using the triple immunosuppressive therapy of tacrolimate corticosteroid and corticosteroid. The concentration / dose of tacrolimus was calculated. The MDR1 gene was detected by PCR-RFLP. The effects of different 699GA genotypes of MDR1 gene 3435CTC LCO1B3 on the ratio of tacrolimus concentration to dose of tacrolimus were compared. Results there was no significant difference in the ratio of tacrolimus C / D to tacrolimus after different genotypes at the 3435CT locus of the 1: MDR1 gene. There was no significant difference in the ratio of tacrolimus C / D to tacrolimus in the different genotypes of the 699GA locus. Conclusion different genotypes of 699GA of the 10% SLCO1B3 gene have an effect on the ratio of tacrolimus C / D after operation, and tacrolimus C / D ratio is higher after AA in mutant homozygotes.
