HoxD10作为抑癌基因在前列腺癌中的研究

【摘要】:目的:研究同源盒基因(HOX genes)在前列腺癌组织中的表达情况,以及其对前列腺癌细胞增殖、凋亡以及迁移力的影响,并探讨其作为前列腺癌一种新的肿瘤标记物的可能性。 方法:1.利用免疫组化实验检测HoxD10在蛋白水平在前列腺癌与良性前列腺组织中表达差异。2.构建sh-HoxD10前列腺癌细胞株LNCaP和PC3。3.通过划痕实验检测HoxD10对前列腺癌细胞迁移的影响。4.通过细胞增殖实验检测HoxD10对前列腺癌细胞增殖力的影响。5.通过细胞周期实验检测HoxD10对前列腺癌细胞细胞生长速度的影响。 结果:1.免疫组化实验证明了前列腺癌组织中HoxD10的表达量要明显低于良性前列腺组织(结果均为P<0.000)。2. HoxD10抑制前列腺癌细胞LNCaP、PC3的迁移力、增殖力,并能降低其生长速度。 结论:1.HoxD10前列腺癌组织中表达下调。2. HoxD10在前列腺癌中可能作为一个新的抑癌基因。
[Abstract]:Aim: to study the expression of homologous box gene (HOX geneses) in prostate cancer tissues and its effects on the proliferation, apoptosis and migration of prostate cancer cells, and to explore the possibility of its application as a new tumor marker for prostate cancer. Method 1: 1. Immunohistochemical assay was used to detect the difference of HoxD10 expression in prostate cancer and benign prostate tissues. 2. 2. Sh-HoxD10 prostate cancer cell lines LNCaP and PC3.3 were constructed. The effect of HoxD10 on the migration of prostate cancer cells was detected by scratch test. The effect of HoxD10 on the proliferation of prostate cancer cells was detected by cell proliferation assay. The effect of HoxD10 on the growth rate of prostate cancer cells was detected by cell cycle assay. The result is 1: 1. The expression of HoxD10 in prostate cancer tissues was significantly lower than that in benign prostate tissues (P < 0. 000. 2). HoxD10 inhibited the migration, proliferation and growth rate of prostate cancer cell line LNCaPtPC3. Conclusion: 1. The expression of HoxD 10 is down-regulated in prostate cancer. HoxD10 may be a new tumor suppressor gene in prostate cancer.
