硫磺和MEIN抑制雄激素非依赖性前列腺癌生长的研究

【摘要】:前列腺癌是全球第六大癌症,也是全球男性第二大癌症。欧美国家前列腺癌发病率高于亚洲国家,发病率在100/10万,是最常见的男性泌尿生殖系统恶性肿瘤之一。而近年来,随着人寿命的延长以及饮食结构的改变,我国前列腺癌的发病率稳步升高,从2002年的1.6/10万到2012年的9.92/10万,增加了5倍。早期前列腺癌细胞生长依赖于雄激素,通过雄激素剥夺法能够有效治疗,而当其进入雄激素抵抗阶段后由于缺乏有效治疗手段,患者的生活和生命都受到很多威胁。因此,寻找用于治疗晚期雄激素非依赖性前列腺癌的药物及手段,成为大家关注的热点。常用的研究雄激素非依赖性前列腺癌细胞系有DU-145,PC-3,22Rv1等,DU-145和PC-3不表达雄激素受体AR,也不分泌PSA;22Rv1的生长不受雄激素调控,但是其能够表达低水平的AR,并分泌少量PSA,生长依然依赖于AR信号通路,这与临床复发性前列腺癌特点一致。硫磺作为药物的入药成分广泛用于各类疾病的治疗中。近年来,中药被用于前列腺癌的治疗中,尤其对于晚期雄激素抵抗阶段的前列腺癌,中医治疗己成为主要的手段。硫磺为中药的入药成分中的一种,曾有报道指出硫磺具有一定的抑癌作用,因此研究硫磺对前列腺癌的抑制作用具有重要意义。MEIN是由乳清蛋白多肽和乳酸杆菌发酵产物组成。其主要成分乳清蛋白多肽作为一种营养补充剂,具有低胆固醇、脂肪和乳糖含量,易消化吸收,具有极高的生物利用价值。已有的病例报道指出,乳清蛋白粉对泌尿生殖系统癌症患者的术后恢复、提高患者的生活水平具有重要作用。本文分两个独立部分,分别探究硫磺和MEIN对雄激素非依赖性前列腺肿瘤生长的影响及其可能机制。
[Abstract]:Prostate cancer is the sixth largest cancer in the world and the second largest in men. The incidence of prostate cancer in Europe and America is higher than that in Asia, with an incidence of 100 to 100, 000, making it one of the most common malignant tumors in the male genitourinary system. In recent years, with the prolongation of life expectancy and the change of dietary structure, the incidence of prostate cancer in China has increased steadily, from 1.6 / 100000 in 2002 to 99.2 / 100000 in 2012, which has increased fivefold. Early prostate cancer cells are dependent on androgen and can be effectively treated by androgen deprivation. However, when they enter androgen resistance stage, their lives and lives are threatened because of the lack of effective treatment. Therefore, searching for drugs and methods for the treatment of advanced androgen-independent prostate cancer has become a hot topic. The commonly used androgen independent prostate cancer cell lines DU-145, PC-3O22Rv1 and so on do not express androgen receptor AR-1, nor do they secrete PSA-22Rv1, which are not regulated by androgen. However, it can express low levels of ARs and secrete a small amount of PSA.The growth is still dependent on AR signaling pathway, which is consistent with the clinical characteristics of recurrent prostate cancer. Sulfur is widely used in the treatment of various diseases. In recent years, traditional Chinese medicine has been used in the treatment of prostate cancer, especially for advanced androgen resistance stage prostate cancer, Chinese medicine treatment has become the main means. Sulfur is one of the traditional Chinese medicine ingredients, it has been reported that sulfur has a certain anti-cancer effect, so it is important to study the inhibitory effect of sulfur on prostate cancer. MEIN is composed of whey protein polypeptide and Lactobacillus fermentation product. As a nutritional supplement, whey protein polypeptide has low cholesterol, fat and lactose content, easy to digest and absorb, and has high bioavailability. It has been reported that whey protein powder plays an important role in postoperative recovery and improvement of living standards of patients with urogenital cancer. This paper is divided into two independent parts to explore the effect of sulfur and MEIN on androgen independent prostate tumor growth and its possible mechanism.
