PTEN和Beclin1在膀胱癌中的表达及相关性研究

【摘要】:背景:膀胱癌是我国泌尿外科临床上最常见的肿瘤之一,并且90%以上为移行细胞癌,目前的一些研究表明,第10号染色体缺失的磷酸酶和张力蛋白同源基因(Phosphatase and tension homology deleted from chromosome 10,PTEN)和自噬基因Beclin1在膀胱移行细胞癌的发生、发展起着重要的作用,PTEN和Beclin1可能是膀胱移行细胞癌的重要标志物和潜在的治疗靶点。目的:检测膀胱癌组织、正常膀胱组织中PTEN和Beclin1的表达情况并结合膀胱癌临床病理特征,分析二者在膀胱癌发生发展中的作用及相关性。方法:收集患者膀胱癌标本52例(经病理复检均为移行细胞癌),另取正常膀胱组织标本10例作为对照,用免疫组织化学检测PTEN和Beclin1在上述组织标本中的表达情况,结合膀胱移行细胞癌相关临床病理因素,研究二者在膀胱移行细胞癌组织中的表达与膀胱移行细胞癌其他生物学特点之间的关系。结果:1.膀胱移行细胞癌组织和正常膀胱组织中PTEN的阳性表达率,二者分别为53.84%和100%,差异显著,有统计学意义(P0.05);2.膀胱移行细胞癌组织和正常膀胱组织中Beclin1的阳性表达率,二者分别为59.62%和100%,差异显著,有统计学意义(P0.05);3.PTEN的表达与组织病理学分级、TNM分期明显相关(P0.05),而与患者性别、年龄无明显相关(P0.05);Beclin1的表达与组织病理学分级、肿瘤TNM分期明显相关(P0.05),而与患者性别、年龄无明显相关(P0.05);4.在膀胱移行细胞癌中,PTEN和Beclin1的阳性表达率呈明显的正相关(r=0.378,P0.05)。结论:1.与正常膀胱组织相比,PTEN基因在膀胱移行细胞癌组织中表达水平明显降低,且与肿瘤组织病理学分级、TNM分期明显相关(P0.05)。2.与正常膀胱组织相比,Beclin1基因在膀胱移行细胞癌组织中表达水平明显降低,且与肿瘤组织病理学分级、TNM分期明显相关(P0.05)。3.膀胱移行细胞癌组织中PTEN与Beclin1的表达存在正相关性;临床上联合检测这两种标志物在膀胱肿瘤中的表达对肿瘤的诊断具有一定的意义。
[Abstract]:Background: bladder cancer is one of the most common tumors in urology in China, and more than 90% are transitional cell carcinoma. Phosphatase and tension homology deleted from chromosome 10PTEN and autophagy Beclin1 are present in transitional cell carcinoma of bladder. PTEN and Beclin1 may be important markers and potential therapeutic targets for bladder transitional cell carcinoma. Objective: to detect the expression of PTEN and Beclin1 in bladder cancer and normal bladder tissues, and to analyze the role of PTEN and Beclin1 in the carcinogenesis and development of bladder cancer. Methods: the expression of PTEN and Beclin1 were detected by immunohistochemistry in 52 cases of bladder cancer (transitional cell carcinoma) and 10 cases of normal bladder tissue as control. Combined with the clinicopathological factors related to transitional cell carcinoma of bladder, the relationship between their expression in transitional cell carcinoma of bladder and other biological characteristics of transitional cell carcinoma of bladder was studied. The result is 1: 1. The positive expression rate of PTEN in transitional cell carcinoma of bladder and normal bladder was 53.84% and 100, respectively. The difference was significant (P0.05). The positive expression rates of Beclin1 in transitional cell carcinoma of bladder and normal bladder were 59.62% and 100, respectively. The difference was significant (P0.05). 3. The expression of PTEN was significantly correlated with the TNM stage of histological grade (P0.05), but with the sex of the patients. There was no significant correlation between age (P0.05) and tumor TNM stage (P0.05), but there was no significant correlation between age and age (P0.05). The positive expression rates of PTEN and Beclin1 in transitional cell carcinoma of bladder were positively correlated (r = 0.378P 0.05). Conclusion 1. Compared with normal bladder tissue, the expression level of PTEN gene in bladder transitional cell carcinoma (TCC) was significantly lower than that in normal bladder tissue, and was significantly correlated with TNM stage of tumor histopathology (P0.05). Compared with normal bladder tissue, the expression of Beclin1 gene was significantly decreased in transitional cell carcinoma of bladder, and was significantly correlated with TNM stage of tumor histopathology (P0.05). There is a positive correlation between the expression of PTEN and Beclin1 in bladder transitional cell carcinoma, and the detection of the expression of these two markers in bladder cancer has some significance in the diagnosis of bladder cancer.
