淫羊藿苷联合胚胎干细胞对造影剂肾病大鼠模型肾上皮细胞凋亡的影响

【摘要】:目的:探讨淫羊藿苷联合胚胎干细胞对造影剂肾病(CIN)大鼠模型肾上皮细胞的作用及其可能的机制,为临床上淫羊藿苷联合应用提供实验依据。方法:选取SD大鼠50只,随机分为对照组、模型组、淫羊藿苷组、干细胞组和联合给药组,每组10只,除对照组外其他各组大鼠尾静脉注射吲哚美辛(INDO)、N-硝基-LO-精氨酸甲酯(L-NAME)和泛影葡胺建立CIN大鼠模型。检测造模前后血肌酐(Scr)值;采用半定量RT-PCR和Western blotting法检测细胞凋亡相关因子p38MAPK和caspase 3mRNA和蛋白的表达,流式细胞术定量检测肾上皮细胞凋亡率。结果:Scr测定,注射造影剂48h后,大鼠Scr值明显升高,提示造模成功。与模型组比较,联合给药组大鼠Scr值明显下降(P0.05)。与模型组比较,淫羊藿苷组、干细胞组和联合给药组大鼠p38MAPK和caspase-3mRNA和蛋白表达水平降低(P0.05);联合给药组大鼠肾上皮细胞凋亡率明显低于淫羊藿苷组和干细胞组(P0.05)。结论:联合给药对CIN模型大鼠的肾上皮细胞有保护作用,其作用机制可能与抑制细胞凋亡通路中相关因子有关。
[Abstract]:Objective: to investigate the effect of icariin combined with embryonic stem cells on renal epithelial cells in (CIN) rats model of contrast medium nephropathy and its possible mechanism, and to provide experimental evidence for clinical application of icariin. Methods: fifty SD rats were randomly divided into control group, model group, icariin group, stem cell group and combined administration group, with 10 rats in each group. The rat model of CIN was established by injecting indomethacin (INDO) (N-nitro-L-arginine methyl ester) (L-NAME) and meglumine diatrizoate into the tail vein of rats except the control group. The levels of serum creatinine (Scr), the expression of apoptosis-related factors p38MAPK, caspase 3mRNA and protein were detected by semi-quantitative RT-PCR and Western blotting, and the apoptosis rate of renal epithelial cells was quantitatively detected by flow cytometry. Results the Scr value of rats increased significantly 48 hours after injection of contrast agent, indicating that the model was successful. Compared with the model group, the Scr value of the combined administration group decreased significantly (P0.05). Compared with model group, the expression of p38MAPK and caspase-3mRNA and protein in Icariin group, stem cell group and combined administration group were significantly lower than those in Icariin group and stem cell group (P0.05), while the apoptosis rate of renal epithelial cells in combined administration group was significantly lower than that in Icariin group and stem cell group (P0.05). Conclusion: combined administration of drugs has protective effect on renal epithelial cells in CIN model rats, and its mechanism may be related to the inhibition of factors related to apoptosis pathway.
