PDGF-D在肾间质纤维化中对大鼠肾成纤维细胞的影响

【摘要】:目的:探讨PDGF-DD/PDGFR-β信号通路激活对大鼠肾成纤维细胞增殖、转化、细胞外基质表达的影响。方法:体外培养正常大鼠肾成纤维细胞(NRK-49F),按照PDGF-DD刺激浓度分为对照组、1ng/ml组、10ng/ml组、50ng/ml组、100ng/ml组,然后根据50ng/ml PDGF-DD刺激不同时间分为对照组、12h组、24h组、48h组,CCK-8方法检测不同PDGF-DD浓度及PDGF-DD作用不同时间对NRK-49F增殖的影响;Real-time PCR及Western blot检测各组PDGFR-β、α-SMA、collagen typeⅠ、FN、MMP-2、TIMP-1 m RNA、蛋白表达变化。结果:与对照组相比,PDGF-DD可明显刺激NRK-49F增殖,呈现剂量依赖性(各浓度组细胞增殖率分别为8.17%,19.17%,31.37%,30.77%,P0.05)及时间依赖性(各时间组细胞增殖率分别为12.70%,25.07%,39.79%,P0.05)。另外,PDGF-DD可剂量依赖性刺激PDGF-βR、α-SMA、collagen typeⅠ、FN、TIMP-1在m RNA水平及蛋白水平上的表达增加,下调MMP-2的m RNA及蛋白表达。结论:PDGF-DD/PDGFR-β信号通路的激活可显著刺激NRK-49F的增殖、转化及细胞外基质的沉积,从而导致肾纤维化。
