4-羟苯维胺酯抑制大鼠膀胱癌的COX-2的表达和诱导肿瘤细胞凋亡的实验研究

【摘要】:目的:使用N-甲基-N-亚硝基脲(N-methyl-N-nitrosourea,MNU)诱导SD大鼠膀胱肿瘤模型,观察4-羟苯维胺酯(4-HPR)的对大鼠膀胱肿瘤的干预效果。方法:70只SD大鼠随机分成对照组(A组)、造模组(B组)、造模处理组(C组、D组、E组),处理组分别予以4-HPR、阿霉素(ADM)、卡介苗(BCG)行膀胱内灌注。给药结束后1周处死大鼠,用免疫组化检测大鼠膀胱肿瘤中COX-2的表达和微血管密度(MVD);用TUNEL法检测凋亡。结果:标本病理结果提示4-HPR、BCG、ADM对MNU诱导肿瘤的发生有明确的干预效果(P0.05)。4-HPR能有效抑制COX-2的表达及微血管的生成并促使肿瘤细胞的凋亡(P0.05)。结论:4-HPR、BCG、ADM对MNU的致癌过程均有干预作用,4-HPR与ADM、BCG的抗癌作用无显著性差异,且其副反应更少。
[Abstract]:Aim: to observe the intervention effect of 4 hydroxybenzimide (4-HPR) on bladder neoplasms in SD rats induced by N methyl N nitrosoureaurea (MNU). Methods 70 SD rats were randomly divided into control group (group A), model group (group B) and model group (group C and D group). The treated group was given 4-HPRand adriamycin (ADM), BCG (BCG) respectively. The rats were killed at 1 week after administration. The expression of COX-2 and microvessel density (MVD);) in bladder tumor were detected by immunohistochemistry and apoptosis was detected by TUNEL method. Results: the histopathological results showed that 4-HPR-BCG-ADM had a definite intervention effect on tumorigenesis induced by MNU (P0.05) .4-HPR could effectively inhibit the expression of COX-2 and the formation of microvessels and promote the apoptosis of tumor cells (P0.05). Conclusion there is no significant difference in the anticancer effect of BCGADM between ADM,BCG and 4-HPR in the carcinogenic process of MNU, and its side effects are less.
