高迁移率蛋白N2对膀胱癌T24细胞生长和裸鼠移植瘤的抑制作用

【摘要】:目的高迁移率蛋白N2(HMGN2)对膀胱癌T24细胞生长和裸鼠移植瘤的抑制作用及其机制的研究。方法诱导表达得到重组蛋白HMGN2;在体外,MTT及流式细胞术检测HMGN2对T24细胞的生长抑制作用;在体内,建立T24细胞裸鼠移植瘤模型,瘤体周边分别注射PBS、HMGN2和顺铂(DDP),检测各组肿瘤的体积及质量并计算抑瘤率。Western blot检测凋亡蛋白Bcl-2及Bax的表达。结果 MTT及流式结果表明,HMGN2抑制了T24细胞的生长且使T24细胞阻滞于细胞周期的S期;裸鼠移植瘤实验中,HMGN2组和DDP组肿瘤体积及瘤重明显小于PBS对照组,差异具有统计学意义(P0.05),两组的抑瘤率分别为25%和23%;Western结果显示,与PBS组相比,HMGN2组和DDP组能明显下调瘤组织内Bcl-2的表达(P0.05),且诱导Bax的表达(P0.05)。结论 HMGN2能明显抑制T24细胞的增殖和裸鼠移植瘤的生长,其作用机制可能与影响细胞周期以及诱导细胞凋亡有关。
[Abstract]:Objective to investigate the inhibitory effect and mechanism of high mobility protein N2 (HMGN2) on the growth of bladder cancer cell line T24 and transplanted tumor in nude mice. Methods Recombinant protein HMGN2; was induced to express in vitro, MTT and flow cytometry were used to detect the inhibitory effect of HMGN2 on the growth of T24 cells. The tumor volume and mass were measured by peripheral injection of PBS,HMGN2 and cisplatin (DDP), and the expression of apoptotic protein Bcl-2 and Bax were detected by. Western blot. Results MTT and flow cytometry showed that HMGN2 inhibited the growth of T24 cells and blocked T24 cells in S phase of cell cycle, the tumor volume and tumor weight in HMGN2 and DDP groups were significantly lower than those in PBS control group. The results showed that compared with PBS group, HMGN2 group and DDP group could significantly down-regulate the expression of Bcl-2 in tumor tissue (P0.05), and induce the expression of Bax (P0.05). Conclusion HMGN2 can significantly inhibit the proliferation of T24 cells and the growth of xenografts in nude mice, which may be related to the influence of cell cycle and the induction of apoptosis.
