肾透明细胞癌与EMT的相关性研究

【摘要】:目的: 观察EMT相关表型分子E-cadherin、N-cadherin蛋白表达在肾透明细胞癌中的变化,探讨EMT与肾透明细胞癌分期、分级之间的相关性。为临床进一步了解肾癌的发生发展及为判断肾癌预后的研究提供理论依据。 方法: 收集肾透明细胞癌组织46例和癌旁正常组织10例制作组织芯片,应用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结(sP)法检测E-cadherin、N-cadherin蛋白表达。 结果: 肾透明细胞癌E-cadherin阳性表达明显降低,同时N-cadherin阳性表达明显升高,E-cadherin与N-cadherin表达呈显著相关性(P0.05); E-cadherin的阳性表达率与肾透明细胞癌的临床分期、组织分化及有无淋巴结转移有关(P<0.05)。而N-cadherin的阳性表达率与肾透明细胞癌的临床分期、组织分化无明显相关性(P>0.05),,但与有无淋巴结转移有相关性(P<0.01)。 结论: 1、ccRCC发生发展过程中存在EMT,EMT参与ccRCC形成过程,并与肾透明细胞癌恶性程度有相关性。 2、N-cadherin的高表达和E-cadherin的低表达与ccRCC的发生发展,淋巴转移有关。 3、N-cadherin的表达与ccRCC的预后有关
[Abstract]:Aim: to observe the expression of E-cadherin protein in renal clear cell carcinoma (RCC) and to investigate the correlation between EMT and stage and grade of renal clear cell carcinoma (RCC). To further understand the occurrence and development of renal cell carcinoma and to predict the prognosis of renal cell carcinoma. Methods: tissue microarray was made in 46 cases of renal clear cell carcinoma and 10 cases of adjacent normal tissues. The expression of E-cadherin protein was detected by immunohistochemical streptavidin-biotin peroxidase (sP) method. Results: the positive expression of E-cadherin was significantly decreased and the expression of N-cadherin was significantly increased in renal clear cell carcinoma. The expression of E-cadherin and N-cadherin was significantly correlated (P0.05). The positive expression rate of E-cadherin was related to the clinical stage, tissue differentiation and lymph node metastasis of renal clear cell carcinoma (P < 0. 05). However, the positive expression rate of N-cadherin was not correlated with clinical stage and histological differentiation of renal clear cell carcinoma (P > 0. 05), but correlated with lymph node metastasis (P < 0. 01). Conclusion: 1 EMT,EMT is involved in the formation of ccRCC and is related to the malignancy of renal clear cell carcinoma. 2the high expression of N-cadherin and the low expression of E-cadherin are related to the occurrence and development of ccRCC and lymphatic metastasis. 3The expression of N-cadherin is related to the prognosis of ccRCC.
