Girdin在膀胱癌和肾癌组织中的表达及其临床意义

【摘要】:目的: 明确Girdin在膀胱癌、肾癌及其正常黏膜中的表达情况,并分析Girdin与膀胱癌和肾癌临床病理分级分期等因素的相关性,进一步探讨Girdin蛋白与膀胱癌和肾癌发生、发展、浸润转移的关系,寻找与膀胱癌和肾癌浸润转移相关的早期预测标志,并为未来的阻断治疗提供理论上的基础。 方法: 1.采用免疫组织化学染色法分别检测80例膀胱癌组织、20例正常膀胱粘膜组织及80例肾癌组织、20例正常肾组织中Girdin的表达情况,并结合临床病理因素进行相关分析。 2.统计学处理:采用统计学软件包SPSS20.0进行数据分析,免疫组化结果中表达强度的比较采用两独立样本的Wilcoxon秩和检验方法进行统计,高表达率的比较采用χ2检验(chi-square),表达强度的相关性分析采用Spearman等级相关检验。 结果: 1. Girdin在膀胱癌组织和正常膀胱粘膜组织中表达强度的差异有显著性(Z=-2.572,P=0.010),高表达率的差异有显著性(χ2=4.558,P=0.033),膀胱癌组织中Girdin的表达明显增强。Girdin的表达强度在患者性别、年龄、病理分级和尿脱落细胞结果组中的差异无显著性,而在病理分期、吸烟组中的差异有显著性(Z=-2.600, P=0.009; Z=-2.672,p=0.008)。Girdin的高表达率在患者性别、年龄、病理分级和尿脱落细胞结果组中的差异无显著性,而在病理分期、吸烟组中的差异有显著性(χ2=4.832, P=0.028;χ2=4.872, P=0.027)。 2. Girdin在肾癌组织和正常肾组织中表达强度的差异有显著性(Z=-2.537,P=0.011),高表达率的差异有显著性(χ2=5.580,P=-0.018),肾癌组织中c-Met的表达明显增强。Girdin的表达强度在患者性别、年龄、病理分级、吸烟和尿脱落细胞结果组中的差异无显著性,而在病理分期组中的差异有显著性(Z=-2.958,P=0.003)。Girdin的高表达率在患者性别、年龄、病理分级、吸烟和远处转移结果组中的差异无显著性,而在病理分期组中的差异有显著性(χ2=5.074,P=0.024)。 结论: 1.膀胱癌组织中Girdin的表达强度和高表达率明显高于正常膀胱粘膜组织,肾癌组织中Girdin的表达强度和高表达率明显高于正常肾组织,提示Girdin可能在膀胱癌及肾癌的发生、发展中起重要作用。 2.膀胱癌组织中Girdin的表达强度和高表达率与病理分期明显相关,Girdin与膀胱癌的侵袭能力有关。 3. Girdin在膀胱癌和肾癌中的表达强度及高表达率均与患者吸烟情况有关,提示吸烟也许能使Girdin高表达,其对膀胱癌和肾癌发生、发展起重要作用。 4. Girdin可作为判断膀胱癌和肾癌生物学行为的重要指标。
[Abstract]:Objective: to investigate the expression of Girdin in bladder carcinoma, renal cell carcinoma and its normal mucosa, and to analyze the correlation between Girdin and clinicopathological grade and stage of bladder cancer and renal cell carcinoma, and to explore the relationship between Girdin protein and the occurrence and development of bladder cancer and renal cell carcinoma. The relationship between invasion and metastasis, search for early predictive markers associated with invasion and metastasis of bladder cancer and renal cell carcinoma, and provide a theoretical basis for future blocking therapy. Methods: 1. Immunohistochemical staining was used to detect the expression of Girdin in 80 cases of bladder cancer, 20 cases of normal bladder mucosa, 80 cases of renal carcinoma and 20 cases of normal kidney respectively. 2. Statistical analysis: statistical software package SPSS20.0 was used to analyze the data, the expression intensity in immunohistochemical results was compared by Wilcoxon rank sum test of two independent samples, and the high expression rate was compared by 蠂 2 test (chi-square). The correlation analysis of expression intensity was performed with Spearman grade correlation test. Results: 1. There was significant difference in the expression intensity of Girdin between bladder cancer tissue and normal bladder mucosa tissue (ZP0. 010), and there was significant difference in high expression rate (蠂 2 + 4. 558% P0. 033).
