【摘要】:在正常机体,血糖受到神经、内分泌和肝脏的严格调控,维持在一个狭窄的生理范围内。但是,外源性葡萄糖的持续应用仍然可以引起正常机体糖代谢紊乱。已有的临床资料和实验研究已经显示了脾脏与胰岛内分泌功能的关系。我们为了探讨持续外源性葡萄糖干预是否影响胰岛β细胞的内分泌功能及脾脏是否也参与了这一过程。运用外科手术的方法切除小鼠全脾。将实验小鼠分为8组,分别以不含葡萄糖的纯水,10%、15%、20%的葡萄糖饮水喂养正常(4组)和脾切除小鼠(4组)3周。测定小鼠空腹血糖和胰岛素水平,计算各组小鼠高血糖发生率;正常对照和脾切除对照小鼠、高血糖小鼠进行胰岛素释放试验、胰岛组织免疫组化双染色、胰腺组织活性氧簇(ROS)测定、胰岛β细胞葡萄糖转运体(GLUT2)表达分析。20%葡萄糖饮水喂养的正常组和脾切除组小鼠有高血糖出现,发生率分别为60%,75%,高血糖小鼠的血糖浓度分别为8.62±1.27,11.75±0.91 mmol/L,两组间血糖存在统计学差异(p0.05);相比于正常对照组和脾切除对照组,高血糖小鼠胰岛素释放能力也显著降低(p0.05),ROS水平升高、GLUT2表达水平降低,并且,胰岛β细胞出现凋亡;在脾切除组高血糖小鼠,上述指标变化更显著。以上结果说明,20%葡萄糖饮水通过增加胰腺组织ROS水平和降低β细胞GLUT-2表达引起胰岛β细胞的凋亡,胰岛素分泌能力的低下,进而导致小鼠高血糖。脾脏对外源性葡萄糖干预引起的胰岛内分泌功能降低具有一定的保护作用。
[Abstract]:In the normal body, blood sugar strictly regulated by nerve, endocrine and liver, within a narrow physiological range. However, the continuous application of exogenous glucose can still cause normal glucose metabolism. The clinical data and the experimental research has shown that the relationship between the spleen and pancreatic endocrine function. In order to endocrine function to explore whether the sustained and spleen exogenous glucose intervention effect of pancreatic beta cells are also involved in this process. Using the method of surgical resection and splenectomy in mice. These results indicated that drinking 20% glucose by increasing the level of ROS in pancreatic tissue and reduced beta cell GLUT-2 expression induced apoptosis of pancreatic beta cells, insulin secretion ability is low, The spleen has a protective effect on the decrease of the endocrine function of the islets caused by exogenous glucose intervention.
