携带LMP-1基因的骨髓间充质干细胞对骨质疏松大鼠治疗效果的研究

【摘要】:目的:观察携带LMP-1基因的骨髓间充质干细胞对骨质疏松大鼠的治疗效果方法:健康三月龄雌性SD大鼠40只,由山西医科大学动物实验中心提供。随机分成4组,每组各10只:1、卵巢切除组(A组):(1)股骨髓腔注射骨髓间充质干细胞组(A1组);(2)股骨髓腔注射携带LMP-1基因的骨髓间充质干细胞组(A2组);(3)股骨髓腔注射生理盐水组(A3组);2、假手术组(B组)。3、各组大鼠在股骨髓腔注射相关物质后第12周分别测量其体重,骨密度,外周血钙、磷浓度,并进行比较。之后再用空气栓塞法处死大鼠,肉眼观察各大鼠股骨大体标本变化情况,HE染色法观察股骨头病理标本组织学变化情况。结果:髓腔内注射相关物质12周时A1、A2、A3组大鼠骨质各项指标均较B组差,但是A组大鼠之间骨质状况A2优于A1,A1优于A3(P0.05)。结论:携带LMP-1基因的骨髓间充质干细胞对骨质疏松大鼠有一定的治疗效果。
[Abstract]:Objective: To observe the therapeutic effect of cells carrying method osteoporosis rat mesenchymal stem LMP-1 gene in bone marrow: 3 month old healthy female SD rats of 40, provided by the animal experimental center of Shanxi Medical University. Randomly divided into 4 groups, 10 rats in each group: 1, ovariectomized group (A group): (1) femoral cavity injection of bone marrow mesenchymal stem cells group (A1 group); (2) femoral medullary cavity injection with LMP-1 gene in bone marrow mesenchymal stem cells group (A2 group); (3) femoral medullary cavity injection saline group (A3 group); 2, sham operation group (group B).3 group the rats in the femoral marrow cavity injection related substances were measured after twelfth weeks, weight, bone density, peripheral blood calcium, phosphorus concentration, and compared. Then the rats were killed by air embolism, each rat femur specimens of femoral head were observed by naked eye observation, pathological specimens histological changes of HE staining method results: intramedullary injection related substances At 12 weeks, all indexes of bone in A1, A2 and A3 groups were all worse than those in B group, but the bone status A2 in A group was superior to A1 and A1 was better than A3 (P0.05). Conclusion: bone marrow mesenchymal stem cells carrying LMP-1 gene have certain therapeutic effect on osteoporotic rats.
