内脂素对2型糖尿病kkay小鼠骨骼肌组织PI3K和GLUT-4的影响

【摘要】:目的观察内脂素(visfatin)对kkay糖尿病小鼠骨骼肌组织磷脂酰肌醇-3-激酶(PI3K)和葡萄糖转运蛋白4(GLUT-4)的影响。方法 C57BL/6J小鼠随机分为普通饲料喂养组(CON)、高脂饲料喂养组(HFD),kkay小鼠随机分为糖尿病模型组(DM)和visfatin治疗组(V)。采用实时荧光定量PCR和Western blot方法测定各组小鼠骨骼肌中PI3K和GLUT-4的mRNA和蛋白表达。结果 V组腹腔注射visfatin后的血糖较注射前下降(P=0.013),差异有统计学意义。DM组PI3K、GLUT-4的mRNA及蛋白表达量较CON组、HFD组降低(P0.05),差异有统计学意义;V组PI3K、GLUT-4的mRNA及蛋白表达量较DM组增加(P0.05),差异有统计学意义。结论内脂素可上调骨骼肌组织PI3K和GLUT-4的表达。
[Abstract]:Objective to observe the effects of visfatin on phosphatidylinositol 3 kinase (PI3K) and glucose transporter 4 (GLUT-4) in skeletal muscle of kkay diabetic mice.Methods C57BL/6J mice were randomly divided into two groups: the normal diet group, the high fat diet group and the high fat diet group.
