Tim-1蛋白表达及其基因多态性与系统性红斑狼疮患者营养功能的相关性

【摘要】:目的探讨Tim-1蛋白表达及其基因多态性与系统性红斑狼疮(SLE)患者营养功能指标的相关性。方法收集126例SLE患者外周血样;采用ELISA法检测血清中人Tim-1蛋白水平;采用PCR-RFLP法检测外周血DNA样本Tim-1基因-416GC和-1454GA多态性;应用免疫比浊法检测血清标本中前白蛋白、铜蓝蛋白、视黄醇结合蛋白的水平;采用电化学发光法检测血清标本中铁蛋白、1,25-二羟维生素D3的水平。结果血清Tim-1蛋白、前白蛋白、铜蓝蛋白、视黄醇结合蛋白、铁蛋白及1,25-二羟维生素D3的浓度分别为(249.7±30.2)pg/m L、(226±42)μg/m L、(363±95)μg/m L、(29.4±13.2)μg/m L、(355±164)ng/m L、(26.4±11.5)ng/m L;-416GC位点上GG基因型占11.9%,GC基因型占57.1%,CC基因型占31.0%;-1454GA位点上GG基因型占67.5%,GA基因型占26.2%,AA基因型占6.3%。-416GC位点不同基因型之间(F=0.575,P=0.564)及-1454GA位点不同基因型之间(F=1.255,P=0.289)Tim-1蛋白浓度均无统计学差异。Tim-1与前白蛋白显著负相关(r=-0.176,P=0.033),与铜蓝蛋白(r=0.205,P=0.014)和1,25-二羟维生素D3(r=0.166,P=0.042)均显著正相关。-1454GA位点GG基因型患者血清中前白蛋白水平显著降低(P=0.027),AA基因型患者血清中1,25-二羟维生素D3水平显著降低(P=0.024)。结论血清Tim-1蛋白水平及Tim-1基因-1454GA位点多态性与SLE患者营养功能指标存在相关性。
[Abstract]:Objective to investigate the relationship between Tim-1 protein expression and gene polymorphism in patients with systemic lupus erythematosus (SLE). Methods Peripheral blood samples were collected from 126 patients with SLE, serum Tim-1 protein levels were detected by ELISA method, Tim-1 gene -416GC and -1454GA polymorphisms in peripheral blood DNA samples were detected by PCR-RFLP method, prealbumin and ceruloplasmin in serum samples were detected by immunoturbidimetric method. The level of retinol binding protein and the level of ferritin 1 / 25-dihydroxyvitamin D _ 3 in serum samples were determined by electrochemiluminescence (ECL). Results Serum Tim-1 protein, prealbumin, ceruloplasmin, retinol binding protein, The concentrations of ferritin and 1-dihydroxyvitamin D _ 3 were 249.7 卤30.2)pg/m / L ~ (226 卤42) 渭 g / mL ~ (63 卤95) 渭 g / m ~ (-1)) 29.4 卤13.2) 渭 g / m ~ (1 / 3) ~ (355) 卤164)ng/m ~ (26.4) 卤11.5)ng/m / L ~ (26.4) 卤11.5)ng/m / L ~ (416GC), respectively. There was no significant difference in the protein concentration between the two genotypes. Tim-1 was negatively correlated with prealbumin, and was significantly negatively correlated with prealbumin, and positively correlated with r0.205P0.014) and 125-dihydroxyvitamin D3HV 0.166P0.042) in the serum of GG genotype patients with GG locus 1454GA. The level of albumin in serum of patients with AA genotype was significantly lower than that of control group (P < 0. 027), and the serum level of vitamin D 3 was significantly lower than that of control group (P < 0. 024). Conclusion the level of serum Tim-1 protein and the polymorphism of Tim-1 gene -1454GA are correlated with the nutritional function of SLE patients.
