阿托伐他汀钙联合替米沙坦对波动性高糖诱导的心肌细胞凋亡的影响

【摘要】:目的:研究阿托伐他汀钙(Atorvastatin,atv)联合替米沙坦(Telmisartan,TEL)对波动性高糖诱导心肌细胞凋亡的影响。方法:心肌细胞在低浓度葡萄糖(正常浓度)环境下贴壁成长,取对数期生长的心肌细胞分为空白对照组A组,波动性高糖组B组,波动性高糖环境下不同药物干预(分别为高浓度阿托伐他汀钙10.0umol/l、高浓度替米沙坦1×10-6mol/l、高浓度的二者联合使用)的C1-C3组(阿托伐他汀钙及替米沙坦浓度均根据查找相关参考文献所定的高浓度)。建立起波动性高糖的损伤模型,以及药物干预后继续培养24h后通过细胞增殖及细胞毒性检测试剂盒(CCK-8)检测心肌细胞的增殖,细胞凋亡检测试剂盒(TUNEL)检测心肌细胞的凋亡。结果:(1)与对照组比较,波动性高糖组及各药物干预组的心肌细胞增殖均减低。(2)与单纯的波动性高糖组比较,阿托伐他汀钙、替米沙坦及二者联合的干预组对心肌细胞的增殖均有保护作用。(3)二者联合的药物干预组与单一的药物干预组比较,二者联合使用组对心肌细胞的凋亡的保护作用更加明显,要优于单一使用任一种药物。讨论:波动性高糖的环境对心肌细胞具有明显的损伤作用,阿托伐他汀钙、替米沙坦及二者联合后均能降低波动性高糖对心肌细胞的损伤作用,且二者联合对心肌细胞的保护作用更加显著,明显优于单一使用任一种药物。
[Abstract]:Aim: to study the effects of Atto vastvastatin calcium and telmisartanol on apoptosis of cardiomyocytes induced by volatile hyperglycemia. Methods: cardiomyocytes grew in low concentration glucose (normal concentration). The cardiomyocytes growing in logarithmic phase were divided into control group A and volatile high glucose group B, respectively. The C1-C3 group (Atto vastatin calcium and telmisartan concentrations) in volatile high glucose environments (high concentration of Atto vastatin calcium 10.0 umoll / l, high concentration of telmisartan 1 脳 10 -6 mol / L, high concentration of both) were based on phase finding. The high concentration determined in the relevant references. The damage model of volatile high glucose was established, and the proliferation of cardiomyocytes was detected by cell proliferation and cytotoxicity test kit CCK-8, and the apoptosis of cardiomyocytes was detected by Tunel after drug intervention for 24 hours. Results compared with the control group, the proliferation of cardiomyocytes in the volatile high glucose group and the drug intervention group was decreased. (2) compared with the volatile high glucose group, Atto vastatin calcium was found in the control group. The protective effect of telmisartan and its combination on cardiomyocyte proliferation was more obvious than that of single drug intervention group, and the protective effect on cardiomyocyte apoptosis was more obvious in the combined drug intervention group than in the single drug intervention group, and the protective effect of the two groups on cardiomyocyte apoptosis was more obvious than that in the single drug intervention group. It is better to use any drug alone. Discussion: fluctuating high glucose environment has obvious damage effect on cardiomyocytes. Atto vastatin calcium, telmisartan and the combination of both can reduce the damage effect of fluctuating high glucose on cardiomyocytes. The protective effect of the two combination on cardiomyocytes was more significant than that of any single drug.
