辛伐他汀与阿仑膦酸钠对去卵巢大鼠骨丢失及力学性能退变干预效果的比较

【摘要】:目的阿仑磷酸钠是目前一线抗骨质疏松药物,辛伐他汀作为降脂类药物因表现出促骨形成作用潜能而备受关注,文中旨在比较辛伐他汀与阿仑膦酸钠对卵巢切除大鼠骨丢失的作用。方法 6月龄SD大鼠32只,随机数字表法分为假手术组、模型组、辛伐他汀组和阿仑膦酸钠组,每组8只。假手术组:只暴露卵巢,不予切除,直接缝合;其余大鼠接收双侧卵巢切除术。模型组术后给予双蒸水;辛伐他汀组:10 mg/m L辛伐他汀溶于双蒸水制成混悬液,给药剂量5 mg/(kg·d)灌胃;阿仑膦酸钠组:25μg/m L阿仑膦酸钠溶液,皮下注射,每周70μg/kg。以上干预措施持续12周,3个月后处死大鼠。ELISA法分析血清Ⅰ型前胶原氨基端前肽(procollagen typeⅠN-terminal propeptide,PINP)、Ⅰ型胶原羧基端交联端肽(cross-linked Ctelopeptides of Type I collagen,ICTP)含量,双能X线法检测大鼠第4腰椎骨密度、骨组织形态计量学分析其松质骨骨量和微观结构,取第5腰椎行压缩试验,分析其最大载荷、弹性模量等生物力学性能。结果血清学指标:假手术组PINP、ICTP血清水平均显著低于其余3组(P0.05),阿仑膦酸钠组ICTP水平低于模型组和辛伐他汀组(P0.05)。骨密度:模型组骨密度[(0.238±0.007)g/cm~2]低于假手术组[(0.276±0.015)g/cm~2]、辛伐他汀组[(0.250±0.014)g/cm~2]、阿仑膦酸钠组[(0.269±0.014)g/cm~2],差异均有统计学意义(P0.05),辛伐他汀组显著低于假手术和阿仑膦酸钠组(P0.05),假手术组与阿仑膦酸钠组比较的差异无统计学意义(P0.05)。骨组织形态计量学分析:模型组骨小梁相对体积[(19.90±1.69)%]显著低于假手术组[(29.03±2.59)%]和阿仑膦酸钠组[(27.05±1.91)%],差异有统计学意义(P0.05),骨小梁分离度[(357.33±26.55)μm]显著高于假手术组[(211.17±16.56)μm]和阿仑膦酸钠组[(252.50±19.35)μm],差异有统计学意义(P0.05)。生物力学性能:模型组最大载荷和弹性模量均显著低于其余各组(P0.05)。结论辛伐他汀和阿仑膦酸钠均可抑制卵巢切除大鼠骨量丢失,两者对该模型生物力学性能的维持作用无显著区别,但后者维持骨密度的作用更加显著。
[Abstract]:Objective Alendronate is a first-line anti-osteoporosis drug. Simvastatin as a lipid lowering drug has attracted much attention because of its potential to promote bone formation. The purpose of this study was to compare the effects of simvastatin and alendronate on bone loss in ovariectomized rats. Methods Thirty-two 6-month-old SD rats were randomly divided into sham-operated group, model group, simvastatin group and alendronate group with 8 rats in each group. Sham operation group: only exposed ovary, no resection, direct suture; the other rats received bilateral ovariectomy. The model group was given double-steamed water after operation, simvastatin group: 10 mg/m / L simvastatin solution dissolved in double-steamed water to make suspension (5 mg/(kg d), alendronate group: 25 渭 g / mL alendronate sodium solution, subcutaneously injected, 70 渭 g / kg / week.
