大鼠铁缺乏时下丘脑、垂体、肾上腺EPOR表达情况的研究

【摘要】:目的:本实验通过建立铁缺乏Wistar大鼠模型,检测大鼠铁缺乏时血液学参数、EPO、生长激素(GH)、胰岛素样生长因子-1(IGF-1)水平,检测大鼠下丘脑、垂体、肾上腺EPOR表达情况,测体重和体长;研究大鼠铁缺乏时大鼠下丘脑、垂体、肾上腺组织的EPOR表达情况;铁缺乏时大鼠血清GH、IGF-1的表达水平;初步探讨大鼠铁缺乏时对下丘脑、垂体、肾上腺EPOR和GH-IGF-1轴的影响。方法:选择4周龄健康Wistar大鼠40只,适应性喂养1周后,随机分为两组,每组20只,实验组饲以低铁饲料,对照组饲以普通饲料。每周定期内眦静脉采血检测大鼠的血液学指标,称体重,测体长。低铁干预四周后内眦静脉取血检测血清中EPO、GH和IGF-1,并取下丘脑、垂体、肾上腺组织做免疫组织化学,检测EPOR的表达情况。结果:(1)4周后实验组大鼠血红蛋白(122.64±9.55 g/L)和铁蛋白(79.77±24.99umol/L)浓度下降,两组差异具有统计学意义(P0.05),造模成功。(2)两组大鼠体重均有增加,缺铁组增幅较小;缺铁组大鼠血清GH、IGF-1明显低于正常对照组,差异具有统计学意义(P0.05)。(3)免疫组织化学结果显示,EPOR在对照组下丘脑中的表达较少,以0分和1分为主,高表达占12.5%;EPOR在实验组中表达亦较低,但1分和2分表达率均高于对照组;高表达率占37.5%,差异具有统计学意义。EPOR在垂体中的表达较为明显,且实验组高表达率高于对照组,差异具有统计学意义。在观察过程中,课题组亦发现EPOR在神经垂体的表达要高于在腺垂体的表达。EPOR在肾上腺皮质和髓质亦均有表达,且实验组高表达率明显高于对照组,差异具有统计学意义。结论:(1)大鼠铁缺乏时血清GH、IGF-1水平降低,抑制GH-IGF-1轴,影响其生长发育。(2)大鼠铁缺乏时下丘脑、垂体、肾上腺EPOR表达增加,以适应铁缺乏导致组织器官缺血缺氧状态,对组织器官具有保护作用。(3)大鼠铁缺乏时EPOR对下丘脑-垂体-肾上腺轴可能起到保护作用。
[Abstract]:Objective: to establish a rat model of iron deficiency (Wistar) and to detect the levels of EPOR in hypothalamus, pituitary, adrenal gland, body weight and body length by detecting the levels of EPOR, growth hormone (GH), growth hormone (GH) and insulin-like growth factor (IGF-1) during iron deficiency in rats. To study the expression of EPOR in hypothalamus, pituitary and adrenal gland of rats with iron deficiency, and the expression level of GH IGF-1 in serum of rats with iron deficiency, and to explore the effect of iron deficiency on the axis of EPOR and GH-IGF-1 in hypothalamus, pituitary, adrenal gland and adrenal gland in rats. Methods: forty 4-week-old healthy Wistar rats were randomly divided into two groups: the experimental group was fed with low iron diet and the control group was fed with common diet. The blood samples were collected from the medial canthus vein every week to detect the hematological indexes of the rats, to weigh and to measure the body length. Serum EPO-GH and IGF-1 were collected from the inner canthus vein and the hypothalamus, pituitary and adrenal tissues were taken for immunohistochemistry to detect the expression of EPO-GH and IGF-1 after four weeks of low iron intervention. Results after 4 weeks, the concentrations of hemoglobin and ferritin (79.77 卤24.99umoll / L) decreased in the experimental group (122.64 卤9.55 g / L). The difference between the two groups was statistically significant (P 0.05). The results of immunohistochemistry showed that the expression of EPOR in the hypothalamus of the control group was less than that of the control group. The expression of EPOR in the experimental group was lower than that in the control group, but the expression rate of 1 and 2 points was higher than that of the control group, and the high expression rate of EPOR was significantly higher in the pituitary gland than that in the control group, and the high expression rate of EPOR in the experimental group was higher than that in the control group. The difference is statistically significant. In the course of observation, we also found that the expression of EPOR in neurohypophysis was higher than that in adenohypophysis. EPOR was also expressed in adrenal cortex and medulla, and the high expression rate of EPOR in experimental group was significantly higher than that in control group (P < 0.05). Conclusion the serum GH IGF-1 level in rats with iron deficiency decreased, the GH-IGF-1 axis was inhibited, and the growth and development of GH IGF-1 were inhibited. The expression of EPOR in hypothalamus, pituitary and adrenal gland was increased during iron deficiency in rats, in order to adapt to the ischemia and hypoxia of tissues and organs caused by iron deficiency. EPOR may protect hypothalamus-pituitary-adrenal axis during iron deficiency in rats.
