不同干预疗法对去卵巢骨质疏松大鼠干预作用的对比研究

【摘要】:实验目的:本文采用去卵巢骨质疏松大鼠模型,对比观察了全身垂直振动、跑台运动、金雀异黄酮、雌激素和氯化锂对去卵巢骨质疏松大鼠骨密度、骨微结构、成骨调节相关蛋白表达的影响以及对子宫的副作用,旨在为绝经后骨质疏松的临床防治提供实验参考依据。实验方法:将84只3月龄雌性SD大鼠按体重分层后随机分为假手术组、去卵巢组、振动组、跑台组、金雀异黄酮组、氯化锂组和雌激素组,去卵巢第11周时,开始进行不同干预处理。跑台组每天进行1次时间45min、速度18m/min、坡度5。的跑台训练;振动组每天进行2次15min、频率90HZ、7次/周的振动治疗;金雀异黄酮组每天按体重灌胃1次金雀异黄酮,剂量为1mg/kg体重;氯化锂组每周按体重腹腔注射3次氯化锂,剂量为15mg/kg;雌激素组每周按体重颈部皮下注射3次17β-雌二醇,剂量为25ug/kg.干预处理8周时,于末次处理结束36-48小时内,腹主动脉取血处死各组大鼠,用双能臌骨密度仪检测大鼠离体腰椎骨密度,用micro-CT检测腰椎骨微结构,用721分光光度计检测血清ALP水平,用ELISA检测血清P-catenin水平,用NBT-BCIP染色法检测全骨髓细胞ALP的变化,用Western blot方法检测骨组织和子宫P-GSK-3β、ER-a和ERR-a蛋白表达的变化。实验结果: (1)雌激素组、金雀异黄酮组、跑台组和振动组大鼠体重均显著低于去卵巢组,而氯化锂组差异无显著性。(2)雌激素组和跑台组血清E2水平显著高于去卵巢组,而金雀异黄酮组、氯化锂组和振动组差异无显著性;雌激素组、金雀异黄酮组、跑台组、振动组和氯化锂组大鼠血清ALP水平均显著低于去卵巢组;各组大鼠血清β-catenin水平差异均无显著性。(3)跑台组腰椎离体骨密度显著高于去卵巢组,而雌激素素组、振动组、金雀异黄酮组和氯化锂组差异均无显著性。 (4)氯化锂组大鼠体积骨密度、骨体积分数和骨小梁数目显著高于去卵巢组,骨小梁间隙、骨小梁厚度和结构模型指数无显著变化,而雌激素组、金雀异黄酮组、跑台组和振动组所有检测指标均无显著变化。 (5)雌激素组、振动组和氯化锂组大鼠股骨和子宫P-GSK-3β表达水平均显著高于去卵巢组,而骨髓细胞P-GSK-3β表达水平无显著变化;金雀异黄酮组股骨和骨髓细胞P-GSK-3β表达水平显著高于去卵巢组,而子宫P-GSK-3β表达水平差异无显著性;跑台组股骨、骨髓细胞和子宫P-GSK-3β表达水平均显著高于去卵巢组;(6)雌激素组股骨ER-α表达水平均显著高于去卵巢组,胫骨、第三腰椎、骨髓细胞和子宫ER-α表达水平均显著低于去卵巢组;金雀异黄酮组股骨ER-a表达水平显著高于去卵巢组,胫骨和第三腰椎ER-α表达水平显著低于去卵巢组,而骨髓细胞和子宫ER-α表达水平差异无显著性;氯化锂组股骨ER-α表达水平显著高于去卵巢组,胫骨、第三腰椎和子宫ER-α表达水平显著低于去卵巢组,而骨髓细胞ER-α表达水平差异无显著性;跑台组股骨、第三腰椎和骨髓细胞ER-α表达水平显著高于去卵巢组,而胫骨和子宫ER-α表达水平显著低于去卵巢组;振动组大鼠股骨ER-α表达水平显著高于去卵巢组,胫骨、第三腰椎和子宫ER-α表达水平显著低于去卵巢组,而骨髓细胞ER-α表达水平差异无显著性。 (7)雌激素组股骨、胫骨和子宫ERR-a蛋白表达水平显著低于去卵巢组,而第三腰椎和骨髓细胞ERR-a蛋白表达水平显著高于去卵巢组;金雀异黄酮组股骨和胫骨ERR-a蛋白表达水平显著低于去卵巢组,第三腰椎、骨髓细胞和子宫ERR-a蛋白表达水平显著高于去卵巢组;氯化锂组股骨、胫骨和第三腰椎ERR-a蛋白表达水平显著低于去卵巢组,骨髓细胞ERR-a表达水平显著高于去卵巢组,而子宫ERR-a表达水平差异无显著性;跑台组大鼠股骨、胫骨和子宫ERR-a蛋白表达水平显著低于去卵巢组,第三腰椎和骨髓细胞ERR-a蛋白表达水平显著高于去卵巢组;振动组大鼠胫骨和子宫ERR-a蛋白表达水平显著低于去卵巢组,第三腰椎和骨髓细胞ERR-a蛋白表达水平显著高于去卵巢组,而股骨ERR-a蛋白表达水平差异无显著性。实验结论:跑台运动能增加去卵巢骨质疏松大鼠血清E2水平;跑台运动、振动、金雀异黄酮、氯化锂和雌激素均能降低去卵巢骨质疏松大鼠血清ALP水平;跑台运动能减缓去卵巢骨质疏松大鼠骨密度的下降,而氯化锂能改善去卵巢骨质疏松大鼠的骨微结构:跑台运动、振动、金雀异黄酮、氯化锂和雌激素均部位和组织依赖性地调节去卵巢骨质疏松大鼠组织P-GSK-3β、ER-α和ERR-a的蛋白表达水平;跑台运动、振动、金雀异黄酮和氯化锂均无雌激素样增加去卵巢骨质疏松大鼠子宫重量的副作用。
[Abstract]:Objective : To investigate the effects of whole body vertical vibration , treadmill exercise , genistein , estrogen and lithium chloride on the expression of bone mineral density , bone microstructure , bone - forming regulation - related protein in ovariectomized osteoporotic rats , and to provide experimental reference for the clinical prevention and treatment of postmenopausal osteoporosis .
The vibration group was treated twice a day with a frequency of 90HZ and 7 times per week .
The genistein group was given 1 dose of genistein per day , and the dosage was 1 mg / kg body weight ;
The levels of ALP in isolated lumbar vertebrae were measured by means of micro - CT . The levels of serum ALP were measured by means of micro - CT . The levels of serum ALP were detected by using the method of Western blot . The serum levels of serum E2 in estrogen group , genistein group , treadmill group and vibrating group were significantly higher than those in ovariectomized group , while the difference between estrogen group and serum E2 level was higher than that in ovariectomized group , while the difference between the group of genistein and lithium chloride was not significant .
Serum ALP levels in estrogen group , genistein group , treadmill group , vibration group and lithium chloride group were significantly lower than that in ovariectomized group .
( 4 ) The bone mineral density , bone volume fraction and trabecular number were significantly higher than those in the ovariectomized group , but there were no significant changes in the bone density , bone volume fraction and the structure model index in the estrogen group , the vibration group and the lithium chloride group , and the expression level of P - gsk - 3.beta . in the estrogen group , the vibration group and the lithium chloride group was significantly higher than that in the ovariectomized group .
There was no significant difference in the expression level of P - gsk - 3 in the femoral and bone marrow cells of genistein group than in the ovariectomized group .
There was a significant difference in the expression levels of the femur , bone marrow cells and P - gsk - 3 beta in the treadmill group than in the ovariectomized group .
( 6 ) The level of ER - 伪 in estrogen group was significantly higher than that in ovariectomized group , tibia , third lumbar vertebra , bone marrow cell and ER - 伪 expression level was significantly lower than that in ovariectomized group .
The expression level of ER - a in the femur of genistein group was significantly higher than that in ovariectomized group , and the expression level of ER - 伪 in tibia and third lumbar vertebra was significantly lower than that in ovariectomized group , but there was no significant difference between bone marrow cells and ER - 伪 expression level .
The expression level of ER - 伪 in the femur of the lithium chloride group was significantly higher than that in the ovariectomized group , and the expression level of ER - 伪 in the tibia , the third lumbar vertebra and the uterus was significantly lower than that in the ovariectomized group , but there was no significant difference in the ER - 伪 expression level of the bone marrow cells .
The expression level of ER - 伪 in the femur , the third lumbar vertebra and the bone marrow cells was significantly higher than that in the ovariectomized group , while the expression level of ER - 伪 in the tibia and uterus was significantly lower than that in the ovariectomized group .
( 7 ) The levels of ER - a protein in the femur , tibia and uterus of the estrogen group were significantly lower than that in the ovariectomized group , while the expression level of the third lumbar vertebra and the bone marrow cell ERR - a protein was significantly higher than that in the ovariectomized group .
The expression of ERR - a protein in femur and tibia of genistein group was significantly lower than that in ovariectomized group , third lumbar vertebra , bone marrow cell and uterus ERR - a protein expression level was significantly higher than that in ovariectomized group .
The expression level of ERR - a protein in the femur , tibia and third lumbar vertebra of the lithium chloride group was significantly lower than that in the ovariectomized group , and the expression level of the ERR - a in the bone marrow cells was significantly higher than that in the ovariectomized group , while the expression level of the ERR - a in the uterus was not significant ;
The expression level of ERR - a protein in the femur , tibia and uterus of treadmill group was significantly lower than that in ovariectomized group , and the expression level of ERR - a protein in the third and third lumbar and bone marrow cells was significantly higher than that in ovariectomized group .
The serum ALP level of ovariectomized osteoporotic rats can be reduced by treadmill movement , vibration , genistein , lithium chloride and estrogen ;
The treadmill exercise can slow down the bone mineral density in ovariectomized osteoporotic rats , while lithium chloride can improve the bone microstructure of ovariectomized osteoporotic rats : treadmill exercise , vibration , genistein , lithium chloride and estrogen are all located and tissue - dependent to regulate the protein expression levels of the ovariectomized osteoporotic rats .
The treadmill exercise , vibration , genistein and lithium chloride have no estrogen - like increase in the side effects of the uterus weight of the ovariectomized osteoporotic rats .
