不同水平血尿酸对大鼠学习记忆能力的影响

【摘要】:目的观察不同水平血尿酸对大鼠学习记忆能力的影响。方法将48只大鼠随机分为对照组、轻度组、中度组、重度组各12只,后三组采用腹腔注射氧嗪酸钾盐的方法制备不同水平的高尿酸血症(HUA)动物模型,对照组大鼠腹腔注射等量生理盐水,均连续注射给药28 d。采用Morris水迷宫试验观察各组大鼠的学习记忆能力,化学发光法检测大鼠血清超氧化物歧化酶(SOD),免疫组织化学法、酶联免疫吸附试验观察大鼠海马区β-淀粉样蛋白1-42(Aβ1-42)表达情况。结果对照组、轻度组、中度组、重度组大鼠潜伏期分别为(19.6±5.7)、(39.1±6.5)、(36.2±5.6)、(33.7±8.2)s,穿越平台次数分别为(3.2±0.8)、(2.0±0.6)、(2.1±0.4)、(2.3±0.5)次,轻度组、中度组、重度组与对照组相比,P均0.01。对照组、轻度组、中度组、重度组大鼠血清SOD活性分别为(403.9±16.3)、(356.9±22.0)、(372.9±15.7)、(382.1±16.2)U/m L,海马组织Aβ1-42表达阳性细胞数分别为(8.5±2.3)、(70.6±20.5)、(47.5±11.2)、(30.7±7.9)个,海马组织Aβ1-42相对表达量分别为(11.1±3.1)、(46.9±6.3)、(40.8±5.3)、(35.6±5.6)pg/mg,两两相比,P均0.05。结论高于正常的不同水平血尿酸均可导致大鼠学习记忆能力下降,但随着血尿酸水平的升高大鼠学习记忆能力下降程度减轻,其机制可能与尿酸的抗氧化及促氧化双重特性有关。
[Abstract]:Objective to observe the effects of different levels of serum uric acid on learning and memory ability in rats. Methods Forty-eight rats were randomly divided into control group, mild group, moderate group and severe group, 12 rats in each group. Rats in control group were intraperitoneally injected with the same amount of normal saline for 28 days. The learning and memory abilities of rats in each group were observed by Morris water maze test. The expression of 尾 -amyloid protein 1-42A 尾 1-42 in hippocampus of rats was detected by chemiluminescence assay, immunohistochemical method and enzyme-linked immunosorbent assay. Results the latencies of the control group, mild group, moderate group and severe group were 19.6 卤5.7 and 39.1 卤6.5, respectively, and the times of crossing the platform were 3.2 卤0.8, 2.1 卤0.4 and 2.3 卤0.5, respectively. Compared with the control group, the latency of the mild group, the moderate group and the severe group were all 0.011.Results there was no significant difference between the mild group, the moderate group and the control group (P > 0.05). In the control group, mild group, moderate group and severe group, the serum SOD activity was 403.9 卤16.3fen, 356.9 卤22.0mg / L, respectively, and the number of A 尾 1-42 positive cells in hippocampus was 70.6 卤20.57.5 卤11.20.70 卤7.9respectively, and the relative expression of A 尾 1-42 in hippocampus was 11.1 卤3.446.30.30.8 卤6.30.8 卤5.336.36.6pgmg / g, respectively, compared with 0.055.57.9in hippocampus, the relative expression of A 尾 1-42 in hippocampus was 11.1 卤3.1 卤6.30.38. 6 卤5.6pgmg / g / g, respectively. Conclusion different levels of serum uric acid above normal can decrease the ability of learning and memory in rats, but with the increase of serum uric acid level, the ability of learning and memory decreases. The mechanism may be related to the antioxidant and pro-oxidation characteristics of uric acid.
