桑酮碱改善2型糖尿病大鼠的糖脂代谢

【摘要】:目的观察桑酮碱对2型糖尿病大鼠糖脂代谢、胰岛素抵抗、脂肪细胞因子的影响及其作用机制。方法 140只大鼠喂以高脂饲料联合链脲佐菌素(STZ)建立2型糖尿病模型,空腹血糖值16.7 mmol/L,大鼠随机分为模型组、二甲双胍(180 mg/kg)组、桑酮碱(73.5、147、294 mg/kg)组,空白组10只,治疗组灌胃给药12周。给药后每2周测定大鼠空腹血糖(FBG);给药12周后,测定糖化血清蛋白(GSP)、糖化血红蛋白(GHb)及血脂指标(TC、TG、HDL-C、LDL-C);放免法测定血清胰岛素水平,并计算胰岛素抵抗指数(HOMR-IR)和胰岛素敏感指数(ISI);酶联免疫吸附实验(ELISA)测定血清脂联素和瘦素水平。结果桑酮碱显著降低2型糖尿病大鼠的空腹血糖、GSP、GHb、TC、TG、LDL-C、瘦素水平和胰岛素抵抗指数,升高HDL-C、脂联素水平和胰岛素敏感指数。结论桑酮碱能改善2型糖尿病大鼠糖脂代谢,是通过改善胰岛素抵抗和调节脂肪细胞因子水平而成的。
[Abstract]:Objective to observe the effects of carnitine on glucose and lipid metabolism, insulin resistance and adipocyte factor in type 2 diabetic rats. Methods 140 rats were fed with high fat diet combined with streptozotocin (STZ) to establish type 2 diabetic model. The fasting blood glucose value was 16.7 mmol / L. The rats were randomly divided into model group, metformin 180 mg / kg group, mulberry alkaloid 73.5147 294 mg / kg group and blank group (10 rats). The treatment group was given intragastric administration for 12 weeks. The fasting blood glucose (FBGG) was measured every 2 weeks after administration, the glycosylated serum protein (GSPN), glycosylated hemoglobin (GHB) and serum lipids were measured 12 weeks after administration, and the serum insulin level was measured by radioimmunoassay. The insulin resistance index (HOMR-IRI) and insulin sensitivity index (ISI) were calculated, and the serum adiponectin and leptin levels were measured by Elisa. Results the levels of leptin, insulin resistance, HDL-C, adiponectin and insulin sensitivity index were significantly decreased in type 2 diabetic rats. Conclusion Mulberry alkaloids can improve glucose and lipid metabolism in type 2 diabetic rats by improving insulin resistance and regulating the level of adipocyte cytokines.
