长链非编码RNA在类风湿性关节炎患者和健康人PBMCs中差异表达及其发病机制

【摘要】:目的探究长链非编码RNA(IncRNA)在类风湿性关节炎(RA)患者外周血单个核细胞(PBMCs)和健康人PBMCs中的差异表达,并对其分析以进一步阐明RA的发病机制。方法选取2016年1-4月在该院风湿免疫科就诊的RA患者22例作为观察组,22例健康人作为对照组。采用Agilent Human IncRNA芯片测定5例RA患者和5例健康人的PBMCs中的IncRNA和mRNA的差异表达;利用GO以及Pathway分析检测出差异表达的IncRNA的功能分布,构建起IncRNA-mRNA的共表达网络,采用Trans-与Cis-预测其中与RA发病相关的IncRNA。结果 IncRNA在RA患者的PBMCs和健康人的PBMCs中共存在1 623条差异表达,mRNA差异表达则有851条。GO、Pathway分析表明,差异表达的mRNA的主要功能是参与蛋白激酶、核苷酸、金属离子的结合和转录调节,还参与B细胞受体信号通路、TNF信号通路的调节。经过Cis-和Trans-分析预测和Real-time PCR验证后找到3个IncRNA可能与RA发病有关,分别为NONHSAT 120696、uc.80+、NONHSAT 031501。结论 IncRNA在RA患者PBMCs和健康人PBMCs中存在差异表达,NONHSAT 120696、uc.80+、NONHSAT 031501 3个IncRNA可能与RA的发病有关。
[Abstract]:Objective to investigate the differential expression of long chain noncoding RNAs (IncRNAs) in peripheral blood mononuclear cells (PBMCs) of patients with rheumatoid arthritis (RA) and healthy PBMCs, and to further elucidate the pathogenesis of RA. Methods 22 RA patients from January to April 2016 were selected as control group. The differential expression of IncRNA and mRNA in PBMCs of 5 RA patients and 5 healthy controls were detected by Agilent Human IncRNA microarray, the functional distribution of differentially expressed IncRNAs was detected by go and Pathway analysis, and the coexpression network of IncRNA-mRNA was constructed. Trans- and Cis- were used to predict the incidence of RA. Results there were 1 623 differentially expressed mRNAs of IncRNA in PBMCs of RA patients and healthy people, while 851 differentially expressed mRNAs showed that the main function of differentially expressed mRNA was involved in protein kinase and nucleotides. The binding and transcriptional regulation of metal ions also participate in the regulation of B cell receptor signaling pathway and TNF signaling pathway. After Cis- and Trans-analysis prediction and Real-time PCR verification, it was found that three IncRNAs might be associated with RA, namely NONHSAT 120696uc.80 and NONHSAT031501. Conclusion there is a differential expression of IncRNA in PBMCs of RA patients and healthy people. NONHSAT 120696uc.80 NoNHSAT 031501 3 IncRNAs may be related to the pathogenesis of RA.
