阈值下577nm微脉冲激光光凝对早期糖尿病大鼠视网膜血管内皮生长因子、神经生长因子和趋化素表达的影响

【摘要】:目的观察阈值下577 nm微脉冲激光光凝对早期糖尿病大鼠视网膜血管内皮生长因子(vascular endothelial growth factor,VEGF)、神经生长因子(nerve growth factor,NGF)和趋化素(Chemerin)表达的影响。方法采用65 mg·kg~(-1)链脲佐菌素对40只Brown Norway大鼠建立糖尿病大鼠模型,选取20只造模成功者为实验鼠,造模后2周右眼采用IQ577 nm激光行阈值下微脉冲激光处理为微脉冲激光组,左眼不做任何处理为对照组。在微脉冲激光光凝后3 d、7 d、14 d、28 d随机各选取5只大鼠,应用实时荧光定量PCR和Western blot方法检测大鼠双眼视网膜VEGF、NGF和Chemerin表达情况。结果对照组VEGF mRNA和蛋白在3 d、7 d、14 d、28 d表达均增加,差异均有统计学意义(均为P0.05);微脉冲激光组VEGF mRNA和蛋白在3 d、7 d、14 d、28 d均较对照组下降,差异l 均有统计学意义(均为P0.05)。对照组3 d、7 d、14 d、28 d NGF mRNA和蛋白均逐渐下降(均为P0.05),7 d NGF比3 d下降,但差异无统计学意义(P0.05);微脉冲激光组NGF mRNA和蛋白在3 d、7 d、14 d、28 d均较对照组上升,且在14 d达到峰值,差异均有统计学意义(均为P0.05)。对照组Chemerin mRNA和蛋白在3 d、7 d、14 d、28 d表达均增加,差异均有统计学意义(均为P0.05);微脉冲激光组Chemerin mRNA和蛋白在3 d、7 d、14 d、28 d均较对照组下降,差异均有统计学意义(均为P0.05)。结论阈值下577 nm微脉冲激光光凝可降低早期糖尿病大鼠VEGF、Chemerin表达,上调NGF表达。
[Abstract]:Objective to investigate the effects of 577nm micropulse laser photocoagulation on the expression of vascular endothelial growth factor (vascular endothelial growth), nerve growth factor (nerve growth) and chemokine (Chemerin) in retina of early diabetic rats. Methods 40 Brown Norway rats were induced by 65 mg kg-1 streptozotocin, and 20 successful rats were selected as experimental rats. The right eye was treated with IQ577 nm laser at threshold for 2 weeks. The left eye was not treated as control group. Five rats were randomly selected at 3 days, 7 days, 14 days and 28 days after laser photocoagulation. The expression of VEGF and Chemerin in the binocular retina of rats was detected by real-time fluorescence quantitative PCR and Western blot. Results the expression of VEGF mRNA and protein in the control group increased on the 3rd day, 7th day, 14th day, the difference was statistically significant (P0.05), and the VEGF mRNA and protein in the micro-pulse laser group decreased at the 14th day of the 3rd day and the 14th day of the control group, the difference was statistically significant (P0.05). In the control group, the d NGF mRNA and protein decreased gradually (P0.05) on the 3rd day, the NGF at 7 days was lower than that on the third day (P0.05), but the difference was not significant (P0.05), the NGF mRNA and protein in the micropulse laser group were higher than those in the control group on the 3rd day after 7 days and the protein on the 14th day, and reached the peak value on the 14th day. The difference was statistically significant (P0.05). In the control group, the expression of Chemerin mRNA and protein increased on the 3rd day, 7th day and 14th day, the difference was statistically significant (P0.05), while the Chemerin mRNA and protein in the micro-pulse laser group decreased on the 14th day after 3 days, which were significantly lower than those in the control group (P0.05). Conclusion 577nm micro-pulse laser photocoagulation can decrease the expression of VEGF Chemerin and up-regulate the expression of NGF in early diabetic rats.
