GDF8及其相关受体在人正常及PCOS卵巢卵泡中的表达

【摘要】:目的探究GDF8及其相关受体在人正常卵巢及PCOS卵巢卵泡中的表达。方法用免疫组化方法染色卵巢组织病理切片标本,使用显微镜评估GDF8及其受体在细胞中的定位、表达强度。结果1.通过免疫组化方法发现GDF8及其受体(ACVR2A、ACVR2B、ALK5)蛋白在正常卵巢及PCOS卵巢的初级卵泡的部分颗粒细胞中有表达,在始基卵泡的颗粒细胞中不表达。2.无论在正常卵巢还是PCOS卵巢的生长卵泡中(从始基卵泡到10mm窦卵泡)中均可看到GDF8及其受体蛋白在卵母细胞中表达。3.GDF8及其受体在正常卵巢或者PCOS卵巢0-2mm,2-5mm,5-10mm窦卵泡的颗粒细胞及卵泡膜细胞中均有表达,并且随着窦卵泡直径的增加,以上蛋白的表达也逐步显著增加。4.无论在正常卵巢还是PCOS卵巢中,在2-5mm及5-1 Omm的窦卵泡中,GDF8及其受体在颗粒细胞中表达显著高于对应的卵泡膜细胞,在0-2mm窦卵泡中两种细胞中表达强度没有显著性差异。5.在正常卵巢的颗粒黄体细胞及卵泡膜黄体细胞中均可观察到GDF8及其受体蛋白的表达,且在颗粒黄体细胞中表达显著较高。6.在2-5mm及5-10mm窦卵泡的颗粒细胞/卵泡膜细胞中,GDF8及其受体蛋白在PCOS卵巢中表达显著高于在正常卵巢的表达;但是在0-2mm窦卵泡中,PCOS卵巢中的表达与正常卵巢中的表达无差异。结论GDF8及其相关功能受体在人类卵巢的生长卵泡及黄体中的差异性表达提示它们可能在卵泡卵母细胞、颗粒细胞、卵泡膜细胞的生长发育过程中起到调节作用。GDF8及其相关功能受体在PCOS卵巢和正常卵巢窦卵泡中的差异性表达提示其在PCOS发病机制中的潜在作用。
[Abstract]:Objective To investigate the expression of GDF8 and its related receptors in human normal ovary and PCOS ovarian follicles.Methods Immunohistochemistry was used to stain ovarian histopathological sections.The localization and expression intensity of GDF8 and its receptors in ovarian cells were evaluated by microscope.Results 1.GDF8 and its receptors (ACVR2A, ACVR2B, ALK5) were detected by immunohistochemistry. White was expressed in some granulosa cells of normal ovary and primary follicles of PCOS ovary, but not in granulosa cells of primordial follicles. 2. GDF8 and its receptors were expressed in oocytes of both normal ovary and growing follicles of PCOS ovary (from primordial follicles to 10 mm sinus follicles). The expression of GDF8 and its receptors in granulosa cells and follicular membrane cells of 0-2 mm, 2-5 mm, 5-10 mm sinus follicles in normal ovaries and PCOS ovaries increased significantly with the increase of the diameter of sinus follicles. 4. GDF8 and its receptors were expressed in granulosa cells of 2-5 mm and 5-1 Omm sinus follicles in normal ovaries and PCOS ovaries. The expression of GDF8 and its receptor protein in granulosa luteal cells and follicular membranous luteal cells of normal ovaries was significantly higher than that in granulosa luteal cells. 6. The expression of GDF8 and its receptor protein was significantly higher in granulosa luteal cells of 2-5mm and 5-10mm sinus follicles. In granulocyte/follicular membrane cells, the expression of GDF8 and its receptor protein in PCOS ovaries was significantly higher than that in normal ovaries, but in 0-2 mm sinus follicles, the expression of GDF8 and its receptor protein in PCOS ovaries was not different from that in normal ovaries. The differential expression of GDF8 and its related functional receptors in PCOS ovary and normal ovarian sinus follicles suggests its potential role in the pathogenesis of PCOS.
