新疆地区汉族人群转录因子7类似物2基因多态性与2型糖尿病的相关性研究

【摘要】:目的:探讨新疆地区汉族人群转录因子7类似物2(TCF7L2)基因rs11196218. rs10749127、rs290487、rs290481多态性与2型糖尿病(T2DM)的相关性。方法:选取2013年11月一2014年2月新疆医科大学第一附属医院收治的汉族T2DM患者164例为T2DM组,另选取同时期体检的健康人群195例为对照组,测量并记录身高、体质量、腹围、血压,检测血糖、糖化血红蛋白(HbA1c)、胰岛素、C肽、血脂、尿酸水平。提取外周DNA,检测TCF7L2基因rs11196218、rs10749127、 rs290487、rs290481基因型及等位基因频率。多因素Logistic回归分析T2DM的影响因素。结果:对照组与T2DM组收缩压(SBP)比较,差异无统计学意义(P0.05);T2DM组患者舒张压(DBP)、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、空腹血糖及HbA 1c较对照组升高,高密度脂蛋白胆固醇(HDL-C)较对照组降低(P0.05)。对照组与T2DM组rs11196218、rs290481基因型和等位基因频率分布,差异均无统计学意义(P0.05);两组rs10749127 TT基因型分布,差异有统计学意义(P0.05),rs290487 CC基因型分布及等位基因频率分布,差异有统计学意义(P0.05)。rs10749127的CC、CT、TT基因型T2DM患者间空腹血糖及尿酸水平比较,差异有统计学意义(P0.05)。rs290487的CC、CT、TT基因型间各指标比较,差异均无统计学意义(P0.05)。多因素Logistic回归分析结果显示:rs290487CC基因型、腰围、体质指数(BMI)、空腹血糖及HbA1c与T2DM有回归关系(P0.05)。结论:TCF7L2基因rs10749127、rs290487与新疆地区汉族人群的T2DM的发病率有关,未发现rs11196218、rs290481与T2DM发病的关联。
[Abstract]:Objective: to study the rs11196218. of transcription factor 7 analogue 2 (TCF7L2) gene in Xinjiang Han population. Association of rs10749127,rs290487,rs290481 polymorphism with type 2 diabetes mellitus (T2DM). Methods: from November 2013 to February 2014, 164 T2DM patients of Han nationality admitted in the first affiliated Hospital of Xinjiang Medical University were selected as T2DM group, and 195 healthy subjects were selected as control group. Height, body mass and abdominal circumference were measured and recorded. Blood pressure, blood glucose, glycosylated hemoglobin (HbA1c), insulin, C-peptide, blood lipids, uric acid levels. Peripheral DNA, was extracted to detect the rs11196218,rs10749127, rs290487,rs290481 genotype and allele frequency of TCF7L2 gene. Multivariate Logistic regression analysis was used to analyze the influencing factors of T2DM. Results: there was no significant difference in systolic blood pressure (SBP) between control group and T2DM group (P0.05). The diastolic blood pressure of (DBP), triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) fasting blood glucose and HbA 1c in T2DM group were higher than those in control group. High density lipoprotein cholesterol (HDL-C) was lower than that of control group (P0.05). The frequency distribution of rs11196218,rs290481 genotype and allele in control group and T2DM group were not significantly different (P0.05). The genotype distribution of rs10749127 TT in the two groups was statistically significant (P0.05), and the distribution of rs290487 CC genotype and allele frequency was statistically significant (P0.05). The CC,CT, of rs10749127 was statistically significant (P0.05). There were significant differences in fasting blood glucose and uric acid levels among T2DM patients with TT genotype (P0.05). There was no significant difference in CC,CT,TT genotypes of rs290487 (P0.05). Multivariate Logistic regression analysis showed that rs290487CC genotype, waist circumference, body mass index (BMI),) fasting blood glucose and HbA1c were correlated with T2DM (P0.05). Conclusion: TCF7L2 gene rs10749127,rs290487 is related to the incidence of T2DM in Xinjiang Han population, and no association between rs11196218,rs290481 and T2DM is found.
