2型糖尿病颈动脉粥样硬化患者血清脑源性神经营养因子与肿瘤坏死因子-α水平相关性研究

【摘要】:目的:通过研究合并颈动脉粥样硬化的2型糖尿病患者血清脑源性神经营养因子(BDNF)、肿瘤坏死因子-α (TNF-α)及外周血BDNF mRNA表达水平的变化,探讨2型糖尿病患者中BDNF、TNF-α与颈动脉粥样硬化的关系及可能机制,以及BDNF与TNF-α的相关性。方法:选取入住我院的2型糖尿病患者100例,根据彩色多普勒超声检查实验对象颈动脉内中膜层厚度将其分为合并颈动脉粥样硬化患者52例,无颈动脉粥样硬化患者48例,并另外选取50例健康者作为对照组,检查实验对象颈动脉内中膜层厚度、血脂、空腹血糖、空腹胰岛功能、糖化血红蛋白,并检测TNF-α、BDNF浓度及外周血BDNF mRNA表达水平;使用单因素方差分析对3组间均数进行比较,使用独立样本t检验对2组间进行比较,使用Pearson相关分析方法分析2型糖尿病合并颈动脉粥样硬化患者血清BDNF与TNF-α相关性。结果:2型糖尿病伴颈动脉粥样硬化组患者血清BDNF浓度、BDNF mRNA表达水平明显低于单纯2型糖尿病组及正常对照组(P0.05)。2型糖尿病伴颈动脉粥样硬化组患者血清TNF-α浓度明显高于单纯糖尿病组及正常对照组(P0.05)。2型糖尿病合并颈动脉粥样硬化组患者血清BDNF与TNF-α水平相关(r=-0.378,P0.05)。结论:血清TNF-α与BDNF水平相关,提示TNF-α与BDNF共同参与了2型糖尿病颈动脉粥样硬化的发病。TNF-a in patientS with type 2 diabeteS mellituS and

To investigate the relationship between BDNF,TNF- 伪 and carotid atherosclerosis in type 2 diabetic patients and its possible mechanism, and the correlation between BDNF and TNF- 伪. Methods: 100 patients with type 2 diabetes mellitus admitted to our hospital were divided into 52 patients with carotid atherosclerosis and 48 patients without carotid atherosclerosis according to the thickness of carotid intima media. In addition, 50 healthy subjects were selected as control group. Carotid intima-media thickness, blood lipid, fasting blood glucose, fasting islet function, glycosylated hemoglobin, TNF- 伪, BDNF concentration and BDNF mRNA expression in peripheral blood were measured. Single factor analysis of variance (ANOVA) was used to compare the mean of the three groups, independent sample t-test was used to compare the two groups, and Pearson correlation analysis was used to analyze the correlation between serum BDNF and TNF- 伪 in patients with type 2 diabetes mellitus and carotid atherosclerosis. Results: the serum BDNF concentration in patients with type 2 diabetes mellitus with carotid atherosclerosis was determined. The level of BDNF mRNA expression was significantly lower in type 2 diabetes mellitus group than in normal control group (P0.05). The serum TNF- 伪 level in type 2 diabetes patients with carotid artery atherosclerosis was significantly higher than that in simple diabetes mellitus group and normal control group (P0.05). P0.05).
