新疆汉族人群TCF7L2基因rs13266634多态性与2型糖尿病相关研究

【摘要】:目的:探讨TCF7L2基因多态性位点rs13266634与新疆省汉族人群2型糖尿病之间的相关性。方法:将研究对象分为糖尿病组(n=116)和对照组(n=126)两组,研究对象均来自于新疆省汉族人群。通过聚合酶链式反应-限制性内切酶片段长度多态性(PCR-RFLP)技术对研究人群(126名健康人的对照组和116名2型糖尿病患者的糖尿病组)的TCF7L2基因rs13266634多态性进行检测,并比较基因型和等位基因频率。结果:糖尿病组和正常对照组年龄、体重指数、甘油三酯、低密度脂蛋白及高密度脂蛋白之间无显著差异性,P均大于0.05;而糖化血红蛋白、收缩压、舒张压、空腹血糖、餐后2小时血糖及总胆固醇之间有显著差异,均P0.05。TCF7L2基因多态性位点rs13266634的基因型在糖尿病组和正常对照组中的分布平率分别为:按TT、CT、CC顺序为20.7%、48.6%、30.6%和28.6%、42.1%、29.4%,两组间差异均无统计学意义(P0.05); TCF7L2基因rs290487多态位点在两组间的基因型分布符合Hardy-Weinberg平衡定律。Logistic回归校正了年龄、体重指数、性别等因素后,TCF7L2基因rs13266634多态性与2型糖尿病的发生无相关性(P0.05)。结论:新疆汉族人群TCF7L2基因rs13266634 T/C多态性可能与2型糖尿病的发生无相关性。
[Abstract]:Objective: to investigate the association between TCF7L2 gene polymorphism rs13266634 and type 2 diabetes mellitus in Han nationality of Xinjiang province. Methods: the subjects were divided into two groups: diabetes mellitus group (n-116) and control group (n-126). All the subjects were from Han nationality in Xinjiang province. Polymerase chain reaction-restriction endonuclease fragment length polymorphism (PCR-RFLP) was used to detect the rs13266634 polymorphism of TCF7L2 gene in 126 healthy controls and 116 diabetic patients with type 2 diabetes. Genotypes and alleles were compared. Results: there was no significant difference in age, body mass index, triglyceride, low density lipoprotein and high density lipoprotein between diabetic group and normal control group (P > 0.05). There were significant differences in glycosylated hemoglobin, systolic blood pressure, diastolic blood pressure, fasting blood glucose, 2 hours postprandial blood glucose and total cholesterol. The distribution rates of rs13266634 genotypes in the diabetic group and the normal control group were as follows: according to the order of TT,CT,CC, rs13266634 was 20.78.60.30.6% and 28.6%, respectively, and the distribution rate of rs13266634 was 42.1% in the diabetic group and in the normal control group, respectively. 29.4 there was no significant difference between the two groups (P0.05); The genotype distribution of rs290487 polymorphism of TCF7L2 gene in the two groups was in accordance with the Hardy-Weinberg equilibrium law. Logistic regression adjusted for age, body mass index, sex and other factors, TCF7L2 gene rs13266634 polymorphism was not associated with the occurrence of type 2 diabetes (P0.05). Conclusion: TCF7L2 gene rs13266634 T / C polymorphism may not be associated with type 2 diabetes in Xinjiang Han population.
