金黄色葡萄球菌显著增加系统性红斑狼疮患者中性粒细胞NETs的产生

【摘要】:目的探讨金黄色葡萄球菌对系统性红斑狼疮(SLE)中性粒细胞胞外诱捕网(NETs)产生的影响及意义。方法收集SLE稳定者22例、活动者20例和健康对照18例,分离中性粒细胞并加金黄色葡萄球菌共培养,检测金黄色葡萄球菌刺激后NETs的主要成分,DNA试剂盒提取测定游离DNA(f DNA)及酶联免疫吸附测定(ELISA)法测上清LL-37,并分析其与SLE临床及实验指标的关系。结果(1)SLE稳定组、活动组、健康对照组3组中性粒细胞培养上清f DNA量3组间差异有统计学意义,SLE稳定组高于活动组和健康对照组(P分别是0.008、0.027),SLE活动组和健康对照组差异无统计学意义(P0.05);LL-37比较无统计学意义(P0.05)。(2)加入金黄色葡萄球菌共培养,培养上清f DNA量的比较差异有统计学意义,SLE活动组、稳定组均高于健康对照组(P0.05),SLE活动组与稳定组无显著性差异(P0.05);LL-37比较无统计学意义(P0.05)。(3)加入金黄色葡萄球菌共培养后,3组f DNA及LL-37量均较刺激前显著增加,差异均有统计学意义(f DNA P分别是0.000、0.000、0.001,LL-37 P均为0.000);培养前后f DNA的差值SLE活动组显著高于稳定组和健康对照组(P0.05),SLE稳定组、健康对照组差异无统计学意义(P0.05);LL-37差值组间比较均无统计学意义(P0.05)。结论金黄色葡萄球菌刺激后,SLE患者NETs产生/释放量显著增加,SLE活动者最显著,金黄色葡萄球菌对SLE疾病活动可能有影响。
[Abstract]:Objective To study the effect and significance of S. aureus on the extracellular trapping net (NEs) of systemic lupus erythematosus (SLE). Methods 22 cases of SLE,20 cases of active and 18 healthy controls were collected, and the main components of NEs after stimulation of S. aureus were detected by co-culture with Staphylococcus aureus. Serum free DNA (f DNA) and enzyme-linked immunosorbent assay (ELISA) were used to determine the relationship between the clinical and experimental indexes of SLE. Results (1) There was no significant difference between the three groups in the control group, the group of active SLE, the control group and the control group (P = 0.008, 0.027), and there was no significant difference between the SLE group and the healthy control group (P <0.05). LL-37 was not statistically significant (P0.05). (2) The results showed that there was no significant difference between the activity of SLE and the stable group (P <0.05), and there was no significant difference between the SLE and the stable group (P0.05). (3) After the co-culture of staphylococcus aureus, the amount of f-DNA and LL-37 was significantly increased in 3 groups (0.000, 0.000, 0.001, LL-37P was 0.000), and the difference of the difference of f DNA before and after culture was significantly higher than that of stable group and healthy control group (P0.05). There was no significant difference between the control group and the control group (P0.05). Conclusion After the stimulation of S. aureus, the generation/ release of the patients with SLE is significantly increased, and the most significant in the patients with SLE, and the effect of S. aureus on the activities of SLE may be affected.
