绝经后骨质疏松环状RNA表达谱研究及潜在分子标志物筛选

研究背景:环状RNAs(circRNAs)是一类丰富的非编码RNA分子,近期研究发现circRNAs参与微小RNAs(miRNAs)介导的转录后调控,发挥miRNA海绵体的作用。前期研究表明循环系统中许多miRNAs表达变化与绝经后骨质疏松症(Postmenopausal Osteoporosis,PMOP)密切相关,然而 miRNA 作用的具体分子机制尚未研究清楚。circRNAs在PMOP中的表达研究也尚属空白。本文旨在探索circRNAs在绝经后骨质疏松症患者中的表达改变,并在人骨髓间充质干细胞(hMSCs)中验证和分析其功能及作为生物标志物的潜能。研究内容:1、绝经后骨质疏松症环状RNAs差异表达谱的研究利用circRNA芯片技术对PMOP患者与对照组外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中的 circRNAs 进行检测并初步分析。2、绝经后骨质疏松症环状RNAs生物信息学分析对芯片检测到差异表达显著的circRNAs进行整体GO分析和KEGG Pathway分析。选择差异表达显著的5个circRNAs进行RT-PCR验...
