中央杏仁核GLP-1受体信号通路的摄食调控研究

目的:中央杏仁核是胰高血糖素样肽1(GLP-1)摄食调控重要作用位点之一,中央杏仁核内GLP-1受体信号通路在摄食和能量代谢调控中发挥着重要生理作用。本研究拟通过荧光免疫组化染色、核团微量注射等方法,观察激活或阻断中央杏仁核内GLP-1信号通路对大鼠摄食影响及潜在机制。方法:选用成年健康雄性Wistar大鼠,体重280-320 g。采用荧光免疫组织化学染色,观察大鼠中央杏仁核中GLP-1受体的分布;采用脑核团置管及中央杏仁核微量注射GLP-1受体激动剂Ex4或GLP-1受体拮抗剂Ex9,观察GLP-1受体信号通路激活或阻断对大鼠摄食量、高脂饮食、高岭土摄入、以及蔗糖和玉米油摄入的影响。结果:(1)大鼠中央杏仁核和尾壳核分别注射6-羧基荧光素(FAM)标记的Ex4(GLP-1受体激动剂),3小时后中央杏仁核内,而不是尾壳核,可见有呈黄色荧光的FAM,结合荧光免疫组化神经元核(NeuN)染色,确认FAM位于中央杏仁核神经元内。提示,GLP-1受体在中央杏仁核内的表达具有位置特异性。(2)中央杏仁核微量注射GLP-1受体激动剂Ex4,大鼠0-20 h累计摄食量显著降低(P<0.05);...
