甲基莲心碱对UV致人皮肤成纤维细胞氧化损伤的影响

【摘要】:目的探讨甲基莲心碱抗光老化的作用机制,为防治皮肤光老化性疾病提供实验依据。方法建立中长波紫外线辐射人皮肤成纤维细胞光老化模型,实验分为对照组、模型组(单纯UVA+UVB照射)和药物组(UVA+UVB照射后加0.4μmol/L,0.8μmol/L,1.6μmol/L甲基莲心碱干预),检测细胞内ROS的水平和SOD和GPx的活力及MDA的含量。结果与模型组相比,甲基莲心碱可以降低ROS水平及MDA含量(P0.05),同时提高SOD及GPx的活力(P0.05)。结论适当浓度的甲基莲心碱能降低光老化细胞内ROS引起的氧化损伤,增加细胞内抗氧化酶的活性,减轻细胞膜脂质过氧化作用,延缓了皮肤光老化的进程。
[Abstract]:Objective to investigate the anti-photoaging mechanism of methylliensine and to provide experimental evidence for the prevention and treatment of skin photoaging diseases. Methods A photoaging model of human skin fibroblasts irradiated by medium and long wavelength ultraviolet radiation was established and divided into two groups. Model group (simple UVA UVB irradiation) and drug group were treated with 0.4 渭 mol 路L ~ (-1) UVB and 0.4 渭 mol / L ~ (0.8) 渭 mol / L ~ (1.6) mol/L, respectively. The level of ROS, the activity of SOD and GPx, and the content of MDA in the cells were measured, and the results were compared with those in the model group. Methylliensine can decrease the level of ROS and the content of MDA and increase the activity of SOD and GPx. Conclusion the suitable concentration of MML can decrease the oxidative damage induced by ROS and increase the activity of antioxidant enzymes in the cells. It can reduce the lipid peroxidation of cell membrane and delay the process of skin photoaging.
