氟化钠对体外培养的人黑素瘤细胞株A375增殖活性影响的研究

【摘要】: 目的:研究不同浓度氟化钠(NaF)对体外培养的人黑素瘤细胞株A375(A375细胞株)增殖活性的影响,探索NaF对人恶性黑色素瘤(MM)的治疗作用。方法:黑素瘤A375细胞株体外培养24h和48h后,用不同NaF(10,100、500、1000、5000、10000、20000与40000umol/L)处理,四甲基偶氮噻唑蓝(MTT)测定NaF对A375细胞株增殖活性的影响,倒置显微镜观察细胞形态学变化。结果:NaF为10 umol/L时,促进黑素瘤细胞的增殖。从100umol/L开始,随着药物浓度的提高和作用时间的延长,黑素瘤细胞的存活率降低,抑制率提高。但10000、20000与40000umol/L三组作用A375细胞24h,对细胞的增殖影响没有区别(p0.05);20000与40000umol/L两组作用48h,对细胞的增殖影响也没有区别(p0.05)。100与500umol/L表现为抑制作用,大于500umol/L则出现细胞毒性作用,表现为细胞数目明显减少,细胞脱壁,变圆变小,胞膜边界不清,胞核消失等变化。结论:NaF对A375细胞株的增殖活性具有双向调节作用,即低浓度促进细胞增殖,高浓度杀伤和抑制细胞增殖。NaF对A375细胞株增殖活性的影响作用呈剂量-时间-效应关系。
[Abstract]:Objective: to study the effect of sodium fluoride (NAF) at different concentrations on the proliferation of human melanoma cell line A375 (A375) in vitro, and to explore the therapeutic effect of NaF on human malignant melanoma. The effects of NaF on the proliferation of A375 cell line were measured with different NAF 10, 10, 100, 500, 000, 500, 000, 000, 000, 000 and 400 mol / L treatments. The morphologic changes of A375 cell line were observed by inverted microscope. Results when NAF was 10 umol/L, the proliferation of melanoma cells was promoted. With the increase of drug concentration and the prolongation of action time, the survival rate of melanoma cells decreased. The inhibition rate of A375 cells was increased in the three groups of 10000mg / L and 40000umolr / L for 24 h, but there was no difference in the proliferation of A375 cells between the two groups (p 0.05 / L and 40000umolr / L) for 48 h, and the effects on the proliferation of A375 cells were not different between the three groups (P 0.05N 路100 and 500umolr / L), but the cytotoxic effects were observed when the cells were larger than 500umolrL. The results showed that the cell number decreased, the cell dewalling, the circle became smaller, the cell membrane boundary was unclear, and the nucleus disappeared. Conclusion the cell proliferation activity of A375 cell line can be regulated by WNAF in both directions, that is, the low concentration of NAF can promote the proliferation of A375 cell line. The effects of high concentration of killing and inhibiting cell proliferation. NAF on the proliferation activity of A375 cell line showed a dose-time effect relationship.
