大鼠皮肤内移植DMSCs前后表皮中P53和PCNA表达的对比研究

【摘要】: 目的:探讨真皮多能干细胞(dermal multipotential stem cells, DMSCs)对大鼠皮肤光老化(skin photoage)是否具有修复作用,为皮肤光老化的治疗提供新的思路。方法:将48只SD大鼠随机分组:正常组8只和光老化模型组40只,采用UVA和UVB紫外灯同时对模型组进行照射(每天2h,连续照射60天)后,随机取8只大鼠采用HE染色法进行皮肤光老化模型鉴定,确定建模成功后随机将模型组大鼠分为治疗组16只和对照组16只。将体外分离培养鉴定的SD乳鼠DMSCs (1×106细胞悬液)多点注射于治疗组大鼠光老化区皮肤内,每两周一次,而对照组用PBS替代处理。分别对正常组、对照组、治疗组第4周、治疗组第8周大鼠皮肤进行取材,采用免疫组织化学染色法检测大鼠表皮中P53和PCNA的表达。结果:经免疫组织化学染色法检测显示:与对照组相比,4周治疗组大鼠表皮中的P53阳性表达无明显变化,两者比较其差异无统计学意义(P0.05);8周治疗组大鼠表皮中的P53阳性表达减弱,两者比较其差异有统计学意义(P0.05);4周和8周治疗组大鼠表皮中的PCNA的阳性表达均有增强,治疗前后比较均有统计学意义(P0.05)。8周治疗组P53和PCNA与正常对照组比较均无统计学差异性(P0.05)。结论:DMSCs大鼠皮肤内注射后可以抑制光老化皮肤P53蛋白表达、增强PCNA的表达,两者的表达与其在正常大鼠表皮中的表达相一致,考虑DMSCs对光老化可能有一定的修复作用。
[Abstract]:Objective: To investigate the effects of dermal multipotent stem cells (dermal multipotential stem cells, DMSCs) on rat skin photoaging (skin photoage) with repair effect, to provide new ideas for the treatment of skin photoaging. Methods: 48 SD rats were randomly divided into normal group of 8 rats and aging model group with 40 rats. Using UVA and UVB UV light irradiation on the model group at the same time (2H per day, continuous 60 days after irradiation), 8 rats were randomly selected using HE skin photoaging model identification staining identified were established and the model rats were divided into treatment group 16 and control group 16 rats. In vitro isolation and identification of SD rat DMSCs (1 * 106 cell suspension) multi point injection in the treatment of rats photoaging skin, once every two weeks, while the control group treated with PBS respectively.
