寻常型银屑病患者皮损中JAM-1和Claudin-7表达的研究

【摘要】： 目的研究紧密连接蛋白JAM-1和claudin-7在寻常型银屑病患者皮损组织中的表达水平,探讨JAM-1蛋白和claudin-7蛋白在寻常型银屑病中可能的发病机制。方法本课题选取25例寻常型银屑病患者作为实验组,15例健康正常人作为对照组,所用研究方法如下： 1)采用免疫组化法(S-P法)检测寻常型银屑病患者皮损组织和正常对照者皮肤组织中JAM-1、claudin-7蛋白的表达水平； 2)应用逆转录聚合酶链反应法(RT-PCR法)检测寻常型银屑病患者皮损组织和正常对照者皮肤组织中JAM-1、claudin-7 mRNA的表达水平。结果 1)寻常型银屑病患者皮损组织中表皮层角质形成细胞膜上JAM-1蛋白和claudin-7蛋白表达阴性,而正常对照组中JAM-1蛋白和claudin-7蛋白高表达； 2)与正常对照相比,寻常型银屑病患者皮损组织中JAM-1mRNA表达显著降低(银屑病患者vs正常对照者：0.38±0.10 vs2.51±0.09,p=0.03),差异具有统计学意义；claudin-7 mRNA表达亦显著下降(银屑病患者vs正常对照者：0.27±0.12 vs 1.83±0.15,p=0.02),差异具有统计学意义。结论寻常型银屑病皮损中JAM-1蛋白和claudin-7蛋白表达明显降低,可能在银屑病的发病中起重要作用。
[Abstract]:Purpose. To investigate the expression of JAM-1 and claudin-7 in psoriasis vulgaris and to explore the possible pathogenesis of JAM-1 and claudin-7 in psoriasis vulgaris. Method. In this study, 25 patients with psoriasis vulgaris were selected as experimental group and 15 healthy persons as control group. 1) the expression of JAM-1claudin-7 protein in the lesions of patients with psoriasis vulgaris and normal controls was detected by immunohistochemical staining (S-P method). 2) reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of JAM-1claudin-7 mRNA in the lesions of psoriasis vulgaris and the normal controls. Results. 1) the expression of JAM-1 protein and claudin-7 protein in epidermal keratinocytes was negative in psoriasis vulgaris, but JAM-1 protein and claudin-7 protein were highly expressed in normal control group. 2) compared with normal control, The expression of JAM-1mRNA in lesions of psoriasis vulgaris patients was significantly lower than that in normal controls (P < 0. 38 卤0. 10 vs2.51 卤0. 09 vs2.51 卤0. 03), and the expression of claudin-7 mRNA in psoriatic patients was significantly lower than that in normal controls (0. 27 卤0. 12 vs 1. 83 卤0. 15p0. 02). Conclusion. The expression of JAM-1 protein and claudin-7 protein in psoriasis vulgaris may play an important role in the pathogenesis of psoriasis.
