微管不稳定蛋白stathmin在慢性瘙痒中的作用

【摘要】:目的:通过炎症性皮肤病患者及干皮性慢性瘙痒小鼠模型,研究微管不稳定蛋白stathmin在慢性瘙痒发病机制中的作用。方法:收集35例炎症性皮肤病引起的慢性瘙痒患者的皮损标本和3例对照皮肤标本。用免疫组化方法检测皮肤活检标本中stathmin的表达;用苏木精-伊红染色法检测患者表皮组织中病理结构的变化。采用丙酮-乙醚-水诱导的干皮性慢性瘙痒小鼠模型,小鼠的皮损、颈段及腰段脊髓和大脑处取材,通过免疫印迹和实时荧光定量PCR去检测AEW模型组与对照组间stathmin的表达差异。结果:在炎症性皮肤病患者中,与正常对照的皮肤相比,IHC显示,stathmin在患者皮损中表皮角质形成细胞、毛囊上皮细胞和免疫炎症细胞中高表达;HE染色显示患者皮损处增厚,角化过度,并伴有炎症性细胞浸润。在干皮性慢性瘙痒小鼠模型中,相对于对照组,免疫印迹显示,stathmin在AEW组皮损中表达上调,实时荧光定量PCR显示,stathmin在AEW组皮损和大脑中表达上调。结论:研究结果初步表明stathmin在慢性瘙痒的机制中发挥作用。Stathmin通过在角质形成细胞、上皮细胞和免疫炎症细胞中的表达影响皮肤屏障功能,促进慢性瘙痒的进程。Stathmin可能是慢性瘙痒治疗的新靶点,对慢性瘙痒的治疗和新药的研发具有重要意义。
[Abstract]:Aim: to study the role of microtubule unstable protein stathmin in the pathogenesis of chronic pruritus in mice with inflammatory dermatosis and dry skin chronic pruritus. Methods: 35 cases of chronic pruritus caused by inflammatory dermatosis and 3 cases of control skin were collected. Immunohistochemical method was used to detect the expression of stathmin in skin biopsy specimens and hematoxylin-eosin staining was used to detect the pathological structure of epidermis. The mice model of chronic dry skin pruritus induced by acetone ether water was used to detect the difference of stathmin expression between the AEW model group and the control group by immunoblotting and real-time fluorescence quantitative PCR. Results: in the patients with inflammatory dermatosis, IHC showed that stathmin in epidermal keratinocytes, hair follicle epithelial cells and immune inflammatory cells showed thickening in the lesions of the patients compared with the normal skin. Hyperkeratosis accompanied by inflammatory cell infiltration. In the dry skin chronic pruritus mice model, the expression of stathmin was up-regulated in the skin lesions of AEW group compared with the control group, and the expression of stathmin was up-regulated in the skin lesions and cerebrum of AEW group by real-time quantitative PCR. Conclusion: the results suggest that stathmin plays a role in the mechanism of chronic pruritus. Stathmin affects the skin barrier function through its expression in keratinocytes, epithelial cells and immune inflammatory cells. Stathmin may be a new target for the treatment of chronic pruritus, which is of great significance for the treatment of chronic pruritus and the development of new drugs.
