银屑病患者骨髓间充质干细胞细胞因子分泌水平及受体表达的研究

【摘要】: [目的]研究银屑病患者骨髓间充质干细胞(Mesenchymal stem cells, MSCs)细胞因子分泌水平及骨髓单个核细胞(Bone marrow mononuclear cells,BMMNCs)细胞因子受体的表达状况,并与银屑病病变程度进行相关性分析,探讨银屑病患者骨髓造血微环境的改变。 [方法]①采用密度梯度离心法分离银屑病患者与对照组骨髓单个核细胞,通过贴壁培养法分离培养骨髓间充质干细胞,用流式细胞仪鉴定细胞表面标志。②采用ELISA法测定骨髓MSCs培养上清液中白细胞介素-8(IL-8)、白细胞介素-3(IL-3)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)及肝细胞生长因子(HGF)的分泌水平。③用流式细胞仪测定骨髓单个核细胞表面白细胞介素-3受体α(IL-3Rα)和肝细胞生长因子受体(HGFR)表达水平。④根据银屑病皮损面积和严重程度的PASI评分法,采用pearson相关分析评估细胞因子分泌水平与银屑病病变程度的相关性。 [结果]①流式测定显示培养的骨髓间充质干细胞高表达CD44、CD71,而HLA-DR及造血细胞表面标志CD45、CD34阴性表达,说明所培养的骨髓MSCs纯度较高;②银屑病患者组骨髓间充质干细胞IL-8、IL-3、HGF分泌水平均显著低于对照组(P0.05),两组间GM-CSF的分泌水平无统计学差异(P0.05);③银屑病患者组单个核细胞表面IL-3Rα、HGFR表达水平显著低于对照组(P0.05);④银屑病患者组细胞因子分泌水平与PASI评分无相关性。 [结论]①银屑病患者骨髓造血微环境存在异常,对银屑病的发病可能起到重要作用。②银屑病患者骨髓MSCs细胞因子分泌水平与病变程度无相关性。
[Abstract]:[objective] to study the levels of cytokines secreted by mesenchymal stem cells, MSCs) and the expression of cytokine receptors in bone marrow mononuclear cells of psoriatic patients, and to analyze their correlation with the degree of psoriasis. To investigate the changes of bone marrow hematopoietic microenvironment in patients with psoriasis. [methods] 1Bone marrow mononuclear cells were isolated from psoriatic patients and control group by density gradient centrifugation. Bone marrow mesenchymal stem cells were isolated and cultured by adherent culture. Identification of Cell Surface Marker .2 the secretion of Interleukin-8 (IL-8), Interleukin-3 (IL-3), granulocyte-macrophage Colony stimulating Factor (GM-CSF) and Hepatocyte growth Factor (HGF) in the supernatant of Bone Marrow MSCs Culture by flow Cytometer Level .3 the expression of interleukin-3 receptor 伪 (IL-3R 伪) and hepatocyte growth factor receptor (HGFR) on the surface of bone marrow mononuclear cells was measured by flow cytometry. 4. The PASI scoring method was used according to the area and severity of psoriatic lesions. The correlation between cytokine secretion and psoriasis was evaluated by pearson correlation analysis. [results] 1 flow cytometry showed that CD44-CD71 was highly expressed in cultured bone marrow mesenchymal stem cells, while HLA-DR and hematopoietic cell surface marker CD45 and CD34 were negative, indicating that the purity of cultured bone marrow MSCs was high. 2the secretion level of IL-8 and IL-3HGF of bone marrow mesenchymal stem cells in psoriatic patients was significantly lower than that of control group (P 0.05). There was no significant difference in the secretion level of GM-CSF between the two groups. The expression of IL-3R 伪 HGFR on the surface of mononuclear cells in psoriatic patients was significantly lower than that in control group. There was no correlation between cytokine secretion and PASI score in psoriatic patients. [conclusion] 1 abnormal hematopoietic microenvironment in bone marrow of psoriatic patients may play an important role in the pathogenesis of psoriasis. 2 there is no correlation between the level of MSCs cytokine secretion and the degree of lesion in psoriatic patients.
