鲍温病组织中HPV感染与微卫星DNA改变的关系

【摘要】:目的分析鲍温病(BD)组织中人乳头瘤病毒(HPV)感染与微卫星(MS)DNA改变的关系。方法收集2011年1月-2015年10月医院收治的30例BD患者的病理组织标本,作为观察组,选取同期外科切除的正常组织标本30份作为对照组,均采用基因芯片技术检测HPV分型,探讨BD组织HPV感染与微卫星DNA改变的关系。结果观察组HPV感染阳性11例,感染率为36.67%,明显高于对照组(P0.05),且观察组检出HPV亚型均为高危型,其中HPV16亚型感染所占比例最高,为63.64%;观察组MS DNA改变检出率为20.00%,明显高于对照组(P0.05),其LOH所占比例高于对照组(P0.05);BD组织与P53连锁的D17S796、D17S926位点共发生LOH 3例,与错配修复基因hMLH1连锁的D3S1612、D3S1029位点发现MSI 1例,LOH 2例;HPV感染11例中,MS DNA改变5例占45.45%,其中HPV16亚型感染中MS DNA改变中MSI占14.29%,LOH占42.36%;HPV58亚型感染中LOH占100.00%;HPV阴性中LOH 1例,占5.26%;HPV阳性组MS DNA改变发生率明显高于HPV阴性组(P0.05)。结论 BD发生与高危型HPV感染及MS DNA改变有一定相关性,P53基因突变可能参与BD发病过程;高危型HPV感染与BD MS DNA改变有一定的关系。
[Abstract]:Objective to analyze the relationship between the infection of human papillomavirus (HPV) and the DNA changes of MSN in BDB. Methods from January 2011 to October 2015, 30 pathological specimens of BD patients were collected as observation group and 30 normal tissues as control group. HPV typing was detected by gene chip technique. To investigate the relationship between BD tissue HPV infection and microsatellite DNA changes. Results the positive rate of HPV infection in the observation group was 36.67, which was significantly higher than that in the control group (P0.05). The HPV subtypes were found to be high risk type in the observation group, and the HPV16 subtype was the highest in the observation group. The detection rate of MS DNA in the observation group was 20.00, which was significantly higher than that in the control group (P0.05A), and the proportion of LOH in the observation group was higher than that in the control group (P 0.05). There were 3 cases of LOH in the D17S796FN D17S926 locus linked to p53 in the control group. The D3S1612mD3S1029 locus linked to the mismatch repair gene hMLH1 showed that 1 case of MSI had Loh 2 cases of HPV-HPV-infected 11 cases, 5 cases had MS DNA change (45.45%), and in the HPV16 subtype infection, MSI DNA change accounted for 14.29% and MSI accounted for 14.29% of MSI in HPV58 subtype infection. LOH accounted for 1 case of HPV-negative in HPV58 subtype infection, and LOH accounted for 1 case of HPV-negative in HPV58 subtype infection. The incidence of MS DNA changes in 5.26% of the patients with HPV-positive was significantly higher than that in the HPV negative group (P 0.05). Conclusion the mutation of p53 gene may be involved in the pathogenesis of BD with high risk HPV infection and the change of MS DNA, and the high risk HPV infection may be related to the change of BD MS DNA.
