扁平苔藓皮损中VEGF、p-Akt、Survivin表达与凋亡的关系

【摘要】: 目的:研究VEGF、p-Akt和Survivin蛋白在扁平苔藓皮损中的表达情况以及与扁平苔藓皮损中细胞凋亡的关系,探讨PI3K/Akt信号通路和细胞凋亡在扁平苔藓发病中的作用。 方法:采用免疫组织化学法检测30例扁平苔藓皮损及30例正常皮肤组织标本中VEGF、p-Akt和Survivin的表达情况和表达部位。采用原位末端标记技术(TUNEL)法检测皮损处细胞凋亡的情况。并对VEGF、p-Akt和Survivin在皮损中的表达水平进行相关性分析。 结果:VEGF、p-Akt和Survivin的表达均以细胞浆着黄色或棕黄色颗粒为阳性,采用积分光密度(integrated optical density, IOD)值的大小表示阳性表达强度。扁平苔藓皮损中VEGF、p-Akt和Survivin的IOD值分别为13.584±1.131、14.405±1.263和10.537±1.212均明显高于正常对照组(P0.01),并且三项指标的表达水平呈正相关(P0.01)。扁平苔藓组皮损处表皮角质形成细胞的凋亡指数(apoptosis index, AI)为72.817±9.234,明显高于正常对照组28.160±3.464(P0.01)。 结论:扁平苔藓皮损表皮角质形成细胞高表达的VEGF可能通过PI3K/Akt信号通路使Survivin的表达增高。PI3K/Akt信号通路可能参与了扁平苔藓的发病。扁平苔藓皮损处同时存在角质形成细胞的过度增殖和凋亡亢进。
[Abstract]:Aim: to investigate the expression of VEGFP-Akt and survivin protein in the lesions of lichen planus and the relationship between the expression of VEGFP-Akt and apoptosis in lichen planus and to explore the role of PI3K / Akt signaling pathway and apoptosis in the pathogenesis of lichen planus. Methods: the expression and location of VEGFP-Akt and survivin in 30 cases of lichen planus lesions and 30 cases of normal skin tissue were detected by immunohistochemical method. In situ end labeling (Tunel) was used to detect apoptosis in lesions. The expression levels of VEGF-Akt and survivin in skin lesions were analyzed. Results the positive expression of the cell cytoplasm was yellow or brown granules, and the intensity of the positive expression was indicated by the value of integrated optical density (integrated optical density,). The IOD values of VEGFP-Akt and survivin in lichen planus lesions were 13.584 卤1.131, 14.405 卤1.263 and 10.537 卤1.212, respectively, which were significantly higher than those in the normal control group (P0.01), and the expression levels of the three indexes were positively correlated (P0.01). The apoptotic index (apoptosis index, AI) of epidermal keratinocytes in the lichen planus group was 72.817 卤9.234, which was significantly higher than that in the normal control group (28.160 卤3.464). Conclusion: the overexpression of VEGF in epidermal keratinocytes of lichen planus may increase the expression of survivin through PI3K / Akt signaling pathway. PI3K / Akt signaling pathway may be involved in the pathogenesis of lichen planus. Excessive proliferation and apoptosis of keratinocytes exist in the lesions of lichen planus.
