HIF-1α和COX-2在银屑病中的表达及相关性研究

【摘要】:第一部分HIF-lα与COX-2在银屑病皮损中的表达及相关性研究 目的:检测HIF-1α和COX-2在寻常型银屑病皮损及正常人皮肤组织中的表达水平,探讨HIF-1α和COX-2在银屑病发病机理中的作用。 方法:详细收集30例寻常型银屑病患者和20例正常人的临床资料,并收集皮损和皮肤组织;(1)用半定量RT-PCR检测银屑病皮损和正常皮肤组织中HIF-1α和COX-2 mRNA表达情况,(2)用Western Blot检测银屑病皮损和正常皮肤组织中HIF-1α和COX-2蛋白表达情况。 结果:HIF-1α和COX-2 mRNA相对表达量在寻常型银屑病组(1.32±0.04和1.21±0.04)的表达显著高于正常对照组(0.49±0.03和0.31±0.03),两组相比差异有统计学意义(P0.05);且HIF-1αmRNA相对表达量与COX-2 mRNA相对表达量呈正相关(r=0.664,P0.05);HIF-1α和COX-2蛋白在寻常型银屑病组(0.76±0.03和0.62±0.03)的表达显著高于正常对照组(0.44±0.05和0.28±0.06),,两组相比差异均有统计学意义(P0.05);且HIF-1α蛋白表达量与COX-2蛋白表达量呈正相关(r=0.601,P0.05)。 结论: HIF-1α和COX-2在银屑病皮损中异常高表达且呈正相关,可能在银屑病的发生发展过程中起着重要作用。 第二部分HIF-1α和COX-2在豚鼠银屑病样模型皮损中的表达及相关 性研究 目的:研究HIF-1α和COX-2在豚鼠银屑病样动物模型皮损表皮中的表达及相关性,探讨其在银屑病发病机制的作用。 方法:用5%普萘洛尔乳剂外涂豚鼠耳背部皮肤诱导银屑病样动物模型,应用免疫组化法检测正常豚鼠表皮和豚鼠银屑病样动物模型皮损表皮中HIF-1α和COX-2表达。 结果:5%普萘洛尔乳剂外涂所致豚鼠银屑病样动物模型皮损中HIF-1α和COX-2表达水平较正常对照组明显增高,差异有统计学意义(P0.01)。 结论:HIF-1α和COX-2在豚鼠银屑病样动物模型皮损的表皮中表达明显增高且呈正相关;不仅在组织病理学而且在分子水平上与人类银屑病皮损基本一致。
[Abstract]:Part I the expression of HIF-l 伪 and COX-2 in psoriatic lesions and their correlation objective: to detect the expression of HIF-1 伪 and COX-2 in skin tissues of psoriasis vulgaris and normal people, and to explore the role of HIF-1 伪 and COX-2 in the pathogenesis of psoriasis. Methods: the clinical data of 30 patients with psoriasis vulgaris and 20 normal subjects were collected in detail. (1) the expressions of HIF-1 伪 and COX-2 mRNA in psoriatic lesions and normal skin tissues were detected by semi-quantitative RT-PCR, and (2) the expression of HIF-1 伪 and COX-2 protein in psoriatic lesions and normal skin tissues were detected by Western Blot. Results the relative expression of 1: HIF-1 伪 and COX-2 mRNA in psoriasis vulgaris group (1.32 卤0.04 and 1.21 卤0.04) was significantly higher than that in normal control group (0.49 卤0.03 and 0.31 卤0.03), and the relative expression of HIF-1 伪 mRNA was positively correlated with the relative expression of COX-2 mRNA (P 0.05). The expression of HIF-1 伪 and COX-2 protein in psoriasis vulgaris group (0.76 卤0.03 and 0.62 卤0.03) was significantly higher than that in normal control group (0.44 卤0.05 and 0.28 卤0.06). Conclusion: the overexpression of HIF-1 伪 and COX-2 in psoriatic lesions is positively correlated and may play an important role in the development of psoriasis. The expression of HIF-1 伪 and COX-2 in the lesions of guinea-pig psoriasis model objective: to study the expression and correlation of HIF-1 伪 and COX-2 in the epidermis of psoriasis model of guinea pig. To explore its role in the pathogenesis of psoriasis. Methods: 5% propranolol emulsion was applied to induce psoriasis animal model. The expression of HIF-1 伪 and COX-2 in normal guinea pig epidermis and lesion epidermis of guinea-pig psoriasis model were detected by immunohistochemistry. Results the expression of HIF-1 伪 and COX-2 in the lesions of guinea pig psoriatic model induced by 5% propranolol emulsion was significantly higher than that in normal control group (P0.01). Conclusion the expressions of 1 / HIF-1 伪 and COX-2 in the epidermis of psoriasis like animal model of guinea pig were significantly increased and positively correlated with those of human psoriatic lesions, not only in histopathology but also at the molecular level.
