ALA联合IPL的光动力疗法对豚鼠光老化模型治疗作用的研究

【摘要】: 目的探讨5-氨基酮戊酸(ALA)联合强脉冲光(IPL)的光动力学疗法(PDT)对豚鼠光老化模型的影响,并摸索合适的ALA治疗浓度。 方法通过模拟日光光源(SSR)照射建立豚鼠光老化模型,照射后分别外用10%或20%ALA联合IPL治疗,每3周一次共3次。以苏木素—伊红染色、Weigert染色、Siriured染色观察真皮细胞外基质(胶原纤维、弹力纤维)的改变,以透射电镜观察真皮超微结构(成纤维细胞、弹力纤维和胶原纤维)的改变,并与未接受SSR照射的同龄阴性对照组及SSR照射后同龄自然恢复组比较。 结果豚鼠在以松香蜂蜡混合液脱毛后接受亚红斑量SSR照射,每周三次,连续照射13周,光照部位真皮浅层出现了典型的“光化性弹力纤维变性”的损害;弹力纤维排列紊乱、断裂及异常弹力纤维样物质沉积;Ⅰ、Ⅲ型胶原纤维断裂,排列紊乱;成纤维细胞线粒体空泡化,粗面内质网稀疏。10%ALA联合IPL治疗三次后可显示改善上述破坏,真皮浅层嗜碱性物质堆积减少,Ⅰ、Ⅲ型胶原纤维和弹力纤维排列趋于整齐,并且多是长纤维,断裂减少。成纤维细胞线粒体空泡化减轻并伴有内质网扩张,其疗效显著优于基质霜联合IPL治疗部位。20%ALA联合IPL治疗虽然也有促进上述损伤后修复的作用,但并没有进一步提高疗效,反而使部分区域“光化性弹力纤维变性”的损害进一步加重,成纤维细胞线粒体空泡化改变普遍且明显。 结论ALA联合IPL的光动力疗法对光老化有一定改善作用,其疗效优于仅用IPL治疗。选择合适的ALA治疗浓度非常关键,豚鼠的动物实验提示10%ALA可能为较合适的浓度。
[Abstract]:Objective to investigate the effect of photodynamic therapy of 5-aminolevulinic acid (ALA) combined with intense pulse light (IPL) on photoaging of guinea pigs, and to explore the appropriate concentration of ALA. Methods the photoaging model of guinea pigs was established by simulated solar light source (SSR) irradiation. After irradiation, 10% or 20%ALA combined with IPL were treated externally for 3 times every 3 weeks. The changes of extracellular matrix (collagen fiber, elastic fiber) and ultrastructure (fibroblast, elastic fiber and collagen fiber) of dermis were observed by Weigert staining with hematoxylin and eosin staining. The results were compared with those of the same age negative control group without SSR irradiation and the natural recovery group of the same age after SSR irradiation. Results Guinea pigs were exposed to suberythema SSR after depilation with rosin wax mixture, three times a week for 13 weeks. The typical "photoelastic fiber denaturation" was observed in the superficial dermis of the illumination area, and the arrangement of elastic fibers was disordered.
