Ezrin和AQP3在皮肤鳞状细胞癌中的表达

【摘要】:目的: 皮肤鳞状细胞癌(squamous cell carcinoma, SCC)是起源于角质形成细胞或附属器鳞状上皮细胞的恶性肿瘤,是最常见的皮肤恶性肿瘤之一,具有侵袭性,易于发生临近或远位转移,尤其转移到重要的器官,从而危及生命。该病的发生、侵袭和转移机制目前仍不十分清楚,因此研究皮肤鳞状细胞癌侵袭和转移的机理,对于肿瘤的预防和治疗具有重要意义。 埃兹蛋白(Ezrin protein, Ezrin)是将细胞膜蛋白与肌动蛋白-细胞骨架连接起来的桥接蛋白,具有维持细胞形态和运动、连接黏附分子及调节信号转导等功能。目前研究发现,其主要通过与肿瘤细胞表面黏附分子发生作用来影响肿瘤的分化、侵袭性和转移;水通道蛋白(aquaporins, AQPs)是一类能快速转运水的膜整合蛋白,皮肤上主要分布的水通道蛋白是水通道蛋白3 (aquaporin-3,AQP3)。目前发现AQP3参与多种生理病理过程,其高表达与恶性肿瘤的侵袭和转移有着密切的联系。 检测Ezrin蛋白,AQP3在皮肤鳞状细胞癌中的表达,探讨它们在皮肤鳞状细胞癌发生、发展中的意义及其与临床病理特征的相关性,并探讨二者的相互关系以及在皮肤鳞状细胞癌发病中所起的作用,以期为皮肤鳞状细胞癌的发病机制提供理论依据。 材料与方法: 收集石蜡包埋的皮肤鳞状细胞癌标本43例,其中高分化16例,中分化15例,低分化12例。对照组为15例正常皮肤。应用免疫组化SP法检测43例皮肤鳞状细胞癌和15例正常皮肤组织中Ezrin蛋白和AQP3的表达水平,用已知的Ezrin和AQP3阳性的皮肤鳞状细胞癌标本做为阳性对照,以PBS代替一抗作为阴性对照;结合临床病理学资料应用SPSS13.0统计软件包进行统计学分析。 结果: Ezrin蛋白在43例皮肤鳞状细胞癌中阳性率为93%,15例正常皮肤组织中Ezrin蛋白阳性表达率为6.67%,两者比较差异有显著性(P0.01);Ezrin蛋白的表达与肿瘤的分化程度有显著相关性,其阳性率随皮肤鳞状细胞癌病理分级的降低而升高,差异有显著性(P0.01);Ezrin蛋白在皮肤鳞状细胞癌中则以细胞质染色或细胞膜、细胞质均着色为主。AQP3在皮肤鳞状细胞癌和正常皮肤中的阳性表达率分别为88.4%和6.67%,差异有显著性(P0.01);在皮肤鳞状细胞癌中AQP3蛋白的表达与肿瘤的分化程度有显著相关性(P0.01),即随皮肤鳞状细胞癌病理分级的增高而升高;AQP3在皮肤鳞状细胞癌中则以细胞质染色或细胞膜、细胞质均着色为主。Ezrin蛋白和AQP3的表达呈显著正相关,有统计学意义(r=0.573,P0.01)。 结论: 1、Ezrin蛋白在皮肤鳞状细胞癌中表达增高,且表达水平在皮肤鳞状细胞癌中与肿瘤的分化程度呈负相关,即分化越差,表达越高。 2、AQP3在皮肤鳞状细胞癌中表达增高,且表达水平在皮肤鳞状细胞癌中与肿瘤的分化程度呈正相关,即分化越高,表达越高。 3、Ezrin蛋白和AQP3在皮肤鳞状细胞癌中的表达呈正相关,表明二者在皮肤鳞状细胞癌的发生、发展过程中可能具有协同作用。

It is found that AQP3 is involved in many physiological and pathological processes, and its high expression is related to the invasion and metastasis of malignant tumor. To investigate the expression of Ezrin protein and AQP3 in the squamous cell carcinoma of the skin, to explore their significance in the development of squamous cell carcinoma of the skin and their correlation with clinicopathological features, and to explore the relationship between them and the squamous cell carcinoma of the skin.
